Objective To explore the correlation between children’s Henoch Schonlein purpura and Henoch Schonlein purpura nephritis and HLADRB1*12,HLADRB1*07 and ACE genes in Ningxia area,further explore the etiology and pathogenesis of Henoch Schonlein purpura,in order to provide objective evidence for the prognosis of children with Henoch Schonlein purpura.Methods A total of 62 children with Henoch-Schonlein purpura who were hospitalized in the Department of Pediatrics of General Hospital of Ningxia Medical from January 2017 to August 2017 were selected.Among them,8 cases were related with kidney involvement.Selection of 40 cases of healthy physical examination in the same period.The age and gender of the two groups of children had no statistically significant difference in factors(P>0.05).Use the human genome DNA extraction kit to extract DNA,then PCR amplification,agarose gel electrophoresis and Sanger sequencing were completed.HLADRB1*12,HLADRB1*07 and ACE genes were detected,and genotype and allele frequencies were calculated.Compare the differences in the detection rate,genotype and allele expression of the three genes between the experimental group and the control group with the SPSS 21 statistical software.Results 1.The detection rate of HLA-DRB1*12 gene in children with Henoch-Schonlein purpura was significantly higher than that in the healthy control group(P<0.05),while there was no significant difference in the detection rate between HSP and HSPN children,and the difference was not statisticallysignificant(P>0.05).2.The detection rate of HLA-DRB1*07 gene in children with HSP was significantly lower than that in healthy control groups(P<0.05).The detection rate of HLA-DRB1*07 in children with HSP and HSPN was not significantly different,and the difference was not statistically significant(P>0.05).3.There was no significant difference in the proportion of three genotypes of ACE gene in children with HSP,children with HSPN and healthy control group(P>0.05).Conclusion HLADRB1*12 and HLA-DRB1*07 genes were correlated with HSP children in Ningxia area.Among them,HLA-DRB1*12 was the susceptible gene of HSP and HSPN children in Ningxia,HLA-DRB1*07 was resistance gene.There was no significant correlation between ACE gene and children with HSP and HSPN in Ningxia.
