| Objective:1.The hyperlipidemia model of SD rats was successfully established with specific high fat diet,and the periodontitis model of SD rats was successfully established by the method of periodontal ligature.2.We observed the relationship between hyperlipidemia and periodontitis in SD rats by HE staining,IVIS spectrum CT and other items.3.We applied nuclear medicine to detect the liver and kidney of experimental animals,and observed the relationship between hyperlipidemia and periodontitis in SD rats.Method: 1 Animal groups and modeling: this experiment was divided into four groups: A-normal,B-hyperlipidemia,C-periodontitis,D-hyperlipidemia and periodontitis.1.1 With high-fat feed,B,D two groups were established as hyperlipidemia model,C and D two groups were ligated with 0.20 mm to ligate the left upper jaw second molar of SD rats,and establish periodontitis model.1.2 After successful establishment of animal models,SD rats in A,B,C and D groups were sacrificed at different times,and the maxillary external fixation was performed.Periodontal tissues were detected.IVIS spectrum CT was used for CT scanning and reconstruction,and alveolar bone loss and alveolar bone mineral density were measured.The periodontal tissue of the left upper maxillary second molar of SD rats was stained with HE.1.3 A,B,C,D group SD rats were sacrificed the liver and kidney sections by HE staining,and the detection of nuclear medicine related projects.2 Statistical Analysis: the SPSS20.0 statistics software was used and the T test was carried out.The test standard was bilateral alpha =0.05.P<0.05 has statistical significance.Results: 1 Modeling: total cholesterol B group with high fat when modeling in the 4W(TC),triglyceride(TG),high density lipoprotein(HDL-C),low density lipoprotein(LDL-C)compared with A group were statistically significant(p<0.05);periodontitis group: gingival sulcus bleeding index(Sulcus Bleeding Index SBI),furcation lesions,CT detection index detection in periodontal ligation at 8W A,C,D group of 22 between the three groups,there was significant difference(P<0.05).2 Observation of periodontal tissue HE staining and IVIS spectrum CT showed that the increase of blood lipid can aggravate inflammatory changes in periodontal tissues and aggravate the absorption and destruction of alveolar bone.3 The liver and kidney specimens of SD rats were tested by HE staining and nuclear medicine.3.1 HE staining: partial hepatocytes in group B and group D were fatty degeneration,and the liver of group D was found to be infiltrated with local multiple lymphocytes.In group B and group D,there were a few pathological changes in the small arteries of the kidneys.In group D,local focal lymphocytic infiltration was found.3.2 Serum nuclear medicine detection: compared with group A,HA and CIV increased significantly in group B,which was statistically significant.Compared with group B,LN,HA and CIV increased significantly in D group.Conclusions:1.Using special high fat diet SD rats for 4 weeks,we can successfully establish hyperlipidemia model;8 weeks after application of periodontal ligation,we can successfully establish experimental SD rat model of periodontitis.And provide the basis for the following experiments.2.The degree of periodontitis of the rats with hyperlipidemia and periodontitis was higher than that of rats with periodontitis.It is suggested that hyperlipidemia has an important influence on the occurrence and development of periodontitis.3.The liver and kidney of the rats with hyperlipidemia and periodontitis HE staining suggested that liver and kidney has pathological changes and tissue damage at the same time,the nuclear medicine examination indicated that chronic periodontitis can enhance liver fat storing cell activity in hyperlipidemia rats,weak fibrous tissue degradation,but no obvious effect on the renal function. |
