Objective:To observe intervention effect of classical prescription of shengyangyiwei Decoction on rat model of PLGC NF-?B,Bcl-2,c-myc and Cyclin-D1 expression,and to explore the mechanism of shengyangyiwei Decoction in treating gastric precancerous lesions of the mechanism,provide the basis for clinical use and efficacy theory based shengyangyiwei Decoction in treating PLGC,providing theory the basis for inheriting and exploring the promotion and application of the classic formula of shengyangyiwei decoction.Methods:96 SPF rats,male and female,weight 200±20g,the rats were divided into control group and model group,one week of adaptive feeding,to control group free diet rats,the rats in the MNNG composite modeling method for modeling.The steps are as follows:blank group rats were given free diet the conventional feed,other liquor diluted MNNG model would have been prepared to 150 g/mL free to drink,and will be dissolved at 60 DEG C by intragastric administration of ranitidine brine 2.25g/L,the frequency of once daily dose of 1mL-100g-1.In the sixth week Zaizao module in five rats were randomly selected according to the dose of 5mL/kg given 10%chloral hydrate intraperitoneal anesthesia,after anesthesia,the rats were taken out along the whole stomach,abdominal incision,incision along the greater curvature of the stomach,rinsed with saline,and adsorption of saline water clean stomach tissue residue.Along both sides of the lesser curvature of the stomach removed about0.5-0.8cm wide gastric mucosa in 4%paraformaldehyde fixed,paraffin embedded,sliced,hematoxylin and eosin staining?HE staining?to observe the pathological changes of gastric mucosa in order to determine the success of modeling,if it does not comply with the repeated modeling process.After the success of the model,according to the random number table,the model is divided into model group,positive control group,vitacoenzyme shengyangyiwei Decoction high dose group,shengyangyiwei Decoction middle dose group,low dose group of shengyangyiwei decoction.Then the next step of traditional Chinese medicine and control drug intervention,the way of administration is gavage.After 12 weeks of drug intervention,the rats were killed and the materials were taken for examination.Result:1.Naked eye observation:the rats in the blank group had normal diet,good mental state,sensitive to the external environment,lively and active,fur and luster,and normal weight,and their stools were gray and white.Compared with the blank group rats,the rats in the model group decreased in diet,mental state was poor,reaction was slow,lazy movements were supine,they were fond of getting together,the fur was rough,the weight growth was slow,the body shape was thin,the stool was not formed,and the litter was more severe in the blank group than in the blank group.The diet of the rats in the drug group was better than the model group,the activity increased,the stool formed and the odor was relieved.2.groups of rat gastric mucosa pathological changes:comparison of rat intestinal villus height,crypt depth and villus width:compared with the blank group,model group,intestinal villus height and crypt depth decreased,villus width was significantly increased in model group,significant difference?P<0.01?.Compared with the model group,low dose group of intestinal villus height increased significantly?P<0.05?,positive drug group,middle dose group,high dose group of intestinal villus height increased significantly?P<0.01?;positive group villus width was significantly decreased,the difference was significant?P<0.05?,drug dose group,high dose group of intestinal villus width was significantly decreased,significant difference?P<0.01?,the difference of low drug group intestinal villus width was not significant?P>0.05?;positive drug group,low group crypt depth significantly increased,significant difference?P<0.05?,drug the middle dose group,high dose group significantly increased crypt depth,significant difference?P<0.01?.3.changes in the extent of apoptosis of rats in each group:compared with the blank group,model group,the apoptosis rate was obviously increased,significant difference?P<0.01?;compared with the model group,low dose group apoptosis rate significantly decreased significantly?P<0.05?,positive drug group,middle dose group and high dose group the apoptosis rate significantly decreased significantly?P<0.01?4.the expression of NF-?B,Bcl-2,c-myc and Cyclin-D1 related apoptosis proteins in gastric mucosa of each group was changed:compared with blank group,the expression of NF-?B,Bcl-2,c-myc and Cyclin-D1 protein in model group increased significantly,with significant difference?P<0.01?.Compared with the model group,there was no significant difference in the expression of NF-?B,c-myc and Cyclin-D1protein in the low dose group?P>0.05?,and the expression of Bcl-2 protein in the low dose group was significantly different?P<0.05?.The expression of NF-?B,Bcl-2 and c-myc protein in the positive group,the drug group and the high drug group decreased significantly?P<0.01?.The expression of Cyclin-D1 protein in the positive group was significantly decreased,and the difference was significant?P<0.05?.The expression of Cyclin-D1 protein in the drug group and the high drug group decreased significantly?P<0.01?.5.The expression of gastric mucosa in rats with NF-?B,Bcl-2,c-myc,Bcl-2,Cyclin-D1mRNA groups:compared with the control group,the rats in the model group in general condition is poor,gastric mucosa of rats inNF-?B,Bcl-2,c-myc,Bcl-2,Cyclin-D1mRNA expression increased significantly?P<0.01?;compared with the model group,shengyangyiwei Decoction group rats significantly improved survival,positive group,Chinese medicine high dose group in gastric mucosa of NF-?B,Bcl-2,c-myc,Cyclin-D1 mRNA expression level was significantly decreased?P<0.05?,low dose of Chinese medicine group in gastric mucosa of NF-?B,Bcl-2,c-myc,Cyclin-D1,mRNA expression no significant difference?P>0.05?.Conclusion:shengyangyiwei decoction can regulate the expression of NF-?B,Bcl-2,c-myc,Bcl-2,Cyclin-D1,effective regulation of cell apoptosis,reduce the pathological damage of PLGC lesions of gastric mucosa,improve the digestive function of the stomach,reduce the occurrence and development of gastric mucosa inflammation,which might be a mechanism of shengyangyiwei decoction can inhibit the inflammation of PLGC the development for the shengyangyiwei Decoction Treatment of PLGC provides new theory basis and pharmacodynamics basis.
