ObjectivesTo evaluate the treatment of Inulin on the weight,the fasting blood glucose,the blood lipid,fasting serum insulin and the adipocytokines of metabolic syndrome in rats and explore the influence of PI3 K signal pathway on liver.To study the molecular mechanisms of the PI3 K signal pathway and the relationship between the adipocytokines and this signal pathway.MethodsEighty male Wistar rats aged of 6 weeks divided randomly into normol control group and metabolc syndromn group,Nor group was fed with regular chow,MS group was fed with the high fat diet.At 11 weeks,the rats of MS group were injected by a low dose of STZ(35kg/kg)once,and the metabolic syndrome rats model were stablished.Then the MS rats were randomly divided into four groups: metabolic syndrome group,high dosage group,low dosage group and Rosiglitazone group.And each group was lavaged the corresponding drugs treatments for four weeks.The indexes such as weight,FBG,FINS,lipid,etc,were detected and figuring out the HOMA-IR.Before and after drug treatment the level of FFA,APN,and expression of PI3 K patway of liver in those rats were detected.Results1.Compared with normal group,the rats feeding high glucose,high fat,high salt diet and treated with STZ,the level of weight,FBG,FINS,HOMA-IR,TG,TC and LDL-C was increased and it has statistical significance(P<0.05).2.After treated with the drug,compared with the control group,the weight,FBG,FINS,HOMA-IR,TG and TC has statistical significance(P<0.05).3.After treated with the drug,compared with the control group,the level of FFA and APN has statistical significance(P<0.05).4.After treated with the drug,compared with the control group,the protein expression level of IRS-1,PI3 K,GLUT4 was significantly higher and has statistical significance(P<0.05).5.According to the relative research,there is associativty between APN and IRS-1,FFA and FINS.And there is negative correlation between FFA and IRS-1,APN and FINS.Conclusions1.Inulin can decrease the level of MS rat's weight,FBG,lipid,and the relative Insulin resistance,especially in high dosage group.2.Inulin can improve the level of the adipocytokines in different extent such as FFA,APN and upregulate the protein of IRS-1,PI3 K,GLUT4 in liver of MS rats,and then improved the insulin signal pathway to ameliorate the glucose metabolic and IR,which may be one of the mechanisms to prevent MS.
