Objective: To observe the effects of inulin produced in Gansu on the metabolic syndrome(MS)rats abnormal components,inflammatory factors,oxidative stress and AMPK-ACC/GLUT4 pathway,and explore its molecular mechanism,provide theoretical and experimental basis for the prevention and treatment of MS.Method: The rats model of human metabolic syndrome were replicated by high fat,high sugar,high salt diet and intraperitoneal injection with small dose streptozotocin(STZ).Eighty Specific Pathogen Free male Wistar rats aged 7 weeks were randomly divided into the normal group(n=10)and the experimental group(n=70),which were respectively fed with normal diet and high fat,high sugar,high salt diet.At the time points of the 0,4th,8th,12 th and 16 th weeks,the rats' body weight,abdominal circumference,body length,fasting plasma glucose(FPG),fasting serum insulin(FINS)and systolic and diastolic blood pressure(SBP/DBP)of caudal artery were measured.The rats in experimental group were intraperitoneal injected with streptozocin(STZ)35 mg/kg at the fifteenth week,the effects were evaluated a week later,those were not successful replicated were eliminated,the successful model of rats were randomly divided into 4 groups further: model group(n=10),high dose group(n=10),low dose group(n=10),rosiglitazone group(n=10),each group was lavaged the corresponding drugs treatment,blood pressure and FBG and 2h PG were examined every 2 weeks,the blood lipid,FINS levels were detected and the HOMA-IR was calculated after 8 weeks.ELISA was used to assay the serum level of interleukin 6(IL-6),high sensitive C reactive protein(hs-CRP),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and malondialdehyde(MDA).The liver tissues were stained with HE to observe pathological changes,the expression of GLUT-4 in skeletal muscle was detected by immunohistochemistry,the expression of AMPK?2,p-AMPK?2,ACC? and GLUT4 in corresponding tissues were detected by Western bloting and RT-PCR methods.Results: 1 The rats' body weight,abdominal circumference,SBP,DBP,FBG,2hPG,FINS,HOMA-IR,TG,TC and LDL-C were all significantly higher in the modle group than in the normal group,while HDL-C was lower than that of the normal group,the differences were statistically significant(P<0.05).The above results indicated that the MS rats model were successfully established,it's have the basic characteristics of MS.2 Compared with the model group,after 8 weeks of intervention,the FBG,2h PG,FINS,HOMA-IR,SBP,TG,TC and LDL-C decreased in high dose group,while HDL-C was increased,the differences were statistically significant(P<0.05).3 Compared with the model group,after 8 weeks of intervention,the IL-6,hs-CRP and MDA levels decreased in high dose group,while SOD and GSH-Px levels increased,the differences were statistically significant(P<0.05).4Pathological changes of nonalcoholic fatty liver disease(NAFLD)were observed in model group,while there were a mild pathological changes in high dose group.5 Compared with the normal group,after 8 weeks of intervention,the expression of GLUT-4 in skeletal muscle decreased in mode group detected by immunohistochemistry,while increased in high dose group,the differences were statistically significant(P<0.05).6 Compared with the normal group,after 8 weeks of intervention,the expression of AMPK?2,p-AMPK?2 and GLUT-4decreased in mode group,while ACC? increased,the differences were statistically significant(P<0.05);compared with the model group,the expression of AMPK?2,p-AMPK?2 and GLUT-4 increased in high dose grou,while ACC? decreased,the differences were statistically significant(P<0.05).Conclusions: 1 Inulin produced in Gansu can decrease FBG,2h PG and FINS,improve IR and regulate the lipid metabolism of MS rats,but the function is dose dependenct,high dose group is more effective.2 Inulin produced in Gansu can reduce the inflammation and oxidative stress levels of the MS rats.3 Inulin produced in Gansu have the function of reducing the pathological changes of fatty liver in the MS rats,and prevention the occurrence of nonalcoholic fatty liver disease(NAFLD).4 Inulin produced in Gansu can upregulate the expression of AMPK?2,p-AMPK?2 and GLUT-4,downregulate the expression of ACC?,and regulate the energy metabolism pathway of MS rats,also as AMPK-ACC/GLUT-4 signaling pathway,which may be one of the mechanisms that the Inulin produced in Gansu can prevention and treatment of MS.
