| Objective: In this study,rats were fed with high fat diet to observe whether the lipid lowering effect of GLP-1 was mediated by FGF21/AMPK.Methods:1、40 male SD rats were divided into normal diet group(NC,n=8)and high-fat diet group(HF,n=32),high fat diet group were divided into high-fat diet control group(HF,n=8)and different doses of the drug treatment group,different doses of the drug treatment group and low dose(LL,n=8),middle dose(LM,n=8),high dose(LH,n=8),12 week GLP-1 analogue liraglutide,respectively 50,100,200ug/kg subcutaneous injection,2 times daily for 4 weeks,NC group and HF group were given an equal volume of sterilized saline injection,weekly monitoring rats were weighed and recorded;2 、 using automatic biochemical analyzer for determination to serum triglycerides,total cholesterol,low density lipoprotein cholesterol,high density lipoprotein cholesterol at 12 weeks;3、Western blot was used to detect the expression of FGF21 and AMPK in rat liver tissues at 16 weeks.Results:1、 general situation of ratsAt 16 weeks,measured the weight,compared with HF group,NC group,TC,TG,LDL body weight was significantly increased,HDL decreased,had statistical significance,at 16 weeks,the drug intervention group compared with HF group,body weight,TC,TG,LDL decreased,HDL increased,treatment of low dose intervention group decreased with no statistical significance TC and others had statistical significance.2、 the comparison of the expression level of FGF21 and AMPK in rat liver tissueAccording to the results of Westorn Blot protein analysis,the results of FGF21 protein showed that compared with NC,the expression of HF increased,compared with HF,the expression of FGF21 protein in LL,LM and LH groups was increased,and the difference was statistically significant.The results of P-AMPK protein showed that compared with NC,the expression of protein decreased in HF group.Compared withHF,the expression level of P-AMPK protein in LL,LM and LH groups increased gradually,and the difference was statistically significant.Conclusion:1.GLP-1 analogue liraglutide can reduce body weight and blood lipid.2.GLP-1 analogue liraglutide could raise the level of FGF21 and activation of AMPK signaling pathway. |
