Objective:1.To clarify the effect of Huangqi injection on the survival period and tumor status of mice with advanced hepatocellular carcinoma,and to select the suitable mode of administration for the comprehensive treatment of advanced hepatocellular carcinoma mouse model;2.To monitor the changes of serum VEGF,TNF-? and IL-2 levels in mice under different administration modes,and to investigate the effects of Huangqi Injection on the levels of VEGF,TNF-? and IL-2.Method:In this study,150 mice were selected for the successful model of HCC in kunming,and divided into five groups according to the random number method:A the control group?B chemotherapy group?C chemotherapy + astragalus Injection ? astragalus injection maintenance treatment group?D chemotherapy sequential astragalus injection and astragalus injection maintenance treatment group?E chemotherapy and astragalus injection maintenance treatment group,30 mice per group.The group B,C,D and E underwent chemotherapy at predetermined time,and each 21 days was a cycle of chemotherapy,with a total of 2 cycles.Each mouse was injected with 0.2ml of astragalus injection through the abdominal cavity,once a day.Dynamically monitored the quality of life and the survival period of tumor-bearing mice after treatment began.Observe the changes in the general conditions of tumors in each mouse and plot the tumor growth curve.Four tumor-bearing mice were randomly selected from each group to be dislocated executed on the 21 st,42th,and 63 th days after the start of chemotherapy,removed the tumor tissue completely and weighed it with electronic balance.H.E staining of hepatocellular carcinoma group was conducted and pathological changes of hepatocellular carcinoma were observed.On day 1,21,42,and 63,four tumor-bearing mice were randomly selected from each group.Collected Serum from the tail vein and measured the serum levels of VEGF,TNF-? and IL-2 levels.Results: 1.The significance test of survival rate of mice in each group showed that the differences between group C and group D compared with A were statistically significant(p<0.05);And there was no significant difference between group C and group D(p>0.05),no significant difference between group B and group E compared with group A(p>0.05).2.Mouse tumor size:On the 15 th day after treatment,the difference between group C and group A was statistically significant(p<0.05).On the 30 th day after treatment,the difference between group C and group B was statistically significant(p<0.05).On the 30 th day after treatment,there was a statistically significant difference between group D and group A(p<0.05).On the 42 nd day after treatment,there was a significant difference between group D and group B(p <0.05).On the 42 nd day after treatment,the difference between E group and A group was statistically significant(p<0.05).And there was no significant difference between the groups in other time periods(p > 0.05).3.Weight of mouse tumors:The tumor weight of group A mice was significantly different from that of each treatment group at different time periods(p<0.05).And there was no significant difference between group C and group D(p > 0.05).4.pathological results of mouse tumor cells:There was no obvious necrosis in group A compared with other treatment groups,and the necrotic tumor cells in various treatment groups are more and more delayed with time.By the 63 rd day there was a large area of necrosis,and the most obvious in group C.5.Levels of TNF-?,IL-2,and VEGF:After treatment,serum TNF-? levels in group B mice were significantly different from those in the treatment group(p<0.05).After treatment,serum IL-2 levels in group A mice were significantly different from those in the treatment group(p<0.05).After treatment,the serum VEGF levels in group A were significantly different from those in the treatment group(p<0.05)..Conclusion: 1.In the maintenance treatment of advanced hepatocarcinogenesis mouse model with Huangqi injection,chemotherapy + Huangqi injection ? Huangqi injection maintenance therapy,chemotherapy sequential Huangqi injection ? Huangqi injection maintenance treatment is the best for tumor suppression in mice.Effectively extend the life span of mice and improve the quality of life.2,Huangqi injection can reduce the level of serum VEGF in mice,increase the level of IL-2 and TNF-? to play anti-tumor effect,or serum VEGF,IL-2,TNF-? levels to assess the efficacy of liver cancer.
