Background and Objective:Breast cancer is the major malignant tumor that harms the women's health,whose incidence has been increasing yearly.Triple-negative breast cancer(TNBC)is one type of breast cancer defined as negative for estrogen,progesterone receptor and HER-2,characterized by high degree of malignancy,rapid progress,strong invasion,high recurrence and metastasis rate.Chemoradiotherapy is the main treatment method at present.It's in urgent need to find out the appropriate biological characteristics that can predict the prognosis of TNBC,and thus bring novel therapeutic option for TNBC.This study aims to measure the expression of AR and P53 in TNBC,and inquire into the relationship between the expression of these markers and the prognosis of TNBC.Methods:Clinical data:153 cases diagnosed as TNBC in Qilu Hospital of Shandong University during January 2010 to December 2011 were included in this study.All cases were treated by modified radical mastectomy,confirmed by pathology as invasive ductal carcinoma,without preoperative chemoradiotherapy.The expression of AR and P53 in breast cancer tissues was detected using immunohistochemical staining.The relationship between the expression of these markers and clinicalpathological features was analyzed with?~2 analysis.Survival curves are presented by Kaplan-Meier survival analysis.Cox proportional hazards model was used to achieve the prognostic value of all the individual variables to overall survival(OS)and disease-free survival(DFS).P<0.05 was considered statistically significant.Results:The expression of AR in the 153 cases of triple negative breast cancer was 58.2%,which was significantly associated with lymph node metastasis(P<0.05).While the expression of AR was not correlated to age,tumor size,tumor grade,CK5/6,P63,nerve invasion,vascular invasion,or vascular cancer embolus.The expression of P53 in the cases of TNBC was 60.8%,which is significantly associated with the tumor histological grade(P<0.05),but no association was showed to age,tumor size,lymph node metastasis,CK5/6,P63,nerve invasion,vascular invasion,or vascular cancer embolus.The expressions of AR and P53 were significantly associated with OS and DFS.Tumor size and lymph node metastasis were independent prognostic factors for OS through COX regression analysis.Conlusion:The expression of AR and P53 in TNBC is significantly upgraded,which is associated with lymph node metastasis and histological grade of tumor,respectively.The levels of AR and P53 may be used as important indicators in diagnosis,progress and prognosis of TNBC.
