Objective:To investigate the expression of monoacylglycerol lipase(MAGL)in normal cervix tissue,cervical intraepithelial neoplasia(CIN),cervical squamous carcinoma.The study analyses the relationship between the expression levels of monoacylglycerol lipase and clinical pathological parameters.To explore the role of monoacylglycerol lipase in the occurrence and development of cervical squamous carcinoma and its relationship with the prognosis.Methods:132 specimens of cervical tissue were collected(69 cases of cervical squamous cancer,44 cases of CIN,19 cases of normal cervical tissue)from postoperative cervical tissue in gynecology department of Qingdao University Affiliated Hospital from March 2004 to January 2017.The clinical specimen request patients were not given any preoperative adjuvant radiotherapy or chemotherapy,and were not combined with other cancers.All clinical specimens were obtained bubble in 10% formalin fixed in liquid,and two pathologists reconfirmed the diagnosis of 132 cases.All cervical cancer specimens are squamous cell carcinoma.The clinicopathological parameters were grouped according to age,tumor size,the BMI index,FIGO staging,lymph node metastasis and degree of differentiation.The expression of monoacylglycerol lipase in cervical squamous carcinoma was detected by immunohistochemistry.The relationship between the expression of monoacylglycerol lipase in cervical squamous carcinoma,cervical intraepithelial neoplasia and normal cervical tissue,and the relationship between the expression of monoacylglycerol in cervical squamous carcinoma and the clinicopathological parameters of the patients was analyzed by Chi-square test.Kaplan-Meier method and multivariate COX regression analysis were used to analyze whether MAGL expression and pathological parameters were correlated with the survival of the patient.Results:(1)In normal cervical tissue group,CINI group,CINII-III group and cervical squamous carcinoma group,the positive expression rate of monoacylglycerol lipase was21%?40%?58.3%?75.4%,respectively.There was statistical difference between four groups in the expression of monoacylglycerol lipase(p=0.00,p <0.05).Its expression in normal cervical tissue,CINI group,CINIIIII group and cervical squamous carcinoma expression showed an increasing trend of expression.(2)The positive expression of monacylglycerol lipase in cervical squamous carcinoma was correlated with BMI index,tumor size,FIGO stage and lymph node metastasis(p<0.05),There was no correlation between the expression of MAGL and the age of patients and the degree of tumor differentiation.(p>0.05).(3)The positive expression of monoacylglycerol lipase,BMI index,tumor size,lymph node metastasis and FIGO staging were correlated with the prognosis of patients.Multivariate COX regression analysis showed that FIGO staging and tumor size are independent factors affecting the survival of cervical squamous carcinoma patients.Conclusion:Monoacylglycerol lipase was expressed in normal cervical tissue,cervical CINI,CINIII and cervical squamous carcinoma tissues,and the expression increased progressively,suggesting that monoacylglycerol lipase is related to the development of cervical squamous carcinoma.The positive expression of monoacylglycerol lipase,BMI index,tumor size,FIGO staging,and lymph node metastasis may be correlated with the prognosis of patients.
