Objective:To explore the cause of strong osteogenic ability of ankylosing spondylitis(AS),comparing the osteogenesis ability of the fibroblast cell from ankylosing spondylitis(AS)and lumbar disc herniation(LDH)patients,and the effect of the serum from AS and LDH patients on osteogenesis ability of the fibroblast cell.Methods:The samples were collected from joints and the spine ligament tissue from patients with AS(A fibroblasts)and LDH patients(B fibroblasts).Medium was made with serum of AS(serum A)and LDH patients(serum B).Groups were divided into 4 groups:AA,AB,BA,BB,based on of the fibroblasts(A and B fibroblasts)and serum medium(serum A and B).Phase microscopy observation were taken to observe the osteogenic evolution of fibroblasts.Quantitative and qualitative method were taken to calculate the parameters of alkaline phosphatase(ALP).ELISA was taken to calculate the osteocalcin.Osteogenic ability was compared between AS and LDH based on the parameters of ALP and osteocalcin.Results:Fibroblasts has similar morphological changes in the 4 groups.ALP and osteocalcin expression was increased with the culture,and calculated to evaluate the osteogenic ability.It was found that the expression of ALP and osteocalcin in group AA were larger than those in group AB,and those in group BA were larger than those in group BB,with statistical significance between the four groups(P < 0.05).Conclusion:Fibroblasts from AS and LDH patients have the ability to differentiate into osteoblasts,but fibroblasts from AS have stronger osteogenic ability than LDH.Through the comparison of group AB and BA,it was found serum from AS patients has more powerful to promote bone differentiation,which implies factors in the serum from AS patients may play an important role in the osteogenic differentiation process of fibroblasts.Significance:Fibroblasts from AS patients have stronger osteogenic ability than LDH patients.Genetic factor may play an important role in osteogenic differentiation of fibroblasts.Fibroblasts in the serum of AS patients have stronger osteogenic ability,which implies some factor in AS serum may affect the ability of fibroblast.Further research is needed to find the genetic factor and factor from the AS serum which may explain the osteogenic mechanism of ankylosing spondylitis.
