Background: CD82/KAI1 is specific tumor metastasis suppressor gene,and belongs to four membrane protein superfamily(TM4SF).Signal transductions between cells and extracellular matrix.CD82/KAI1 regulate cell fusion? adhesion?migration and proliferation through a variety of molecular mechanisms.The successful implantation of the embryo involves the complex information exchange between the uterus and the blastocyst.The blastocyst needs to obtain the implantation capacity,while the uterus enters into the "implantation window ".By the activation of mouse blastocysts with endometrial epithelial adhesion reaction,uterine stromal cells began to widely proliferate and differentiate with hyperplasia of decidual area of blood vessels,so the volume and weight of uterus quickly increase,and finally blastocyst is embeded in the uterus of mesangial matrix.The changing process of endometrial is called uterine decidua.According to the preliminary experimental results that KAI1 / CD82 played a certain role in the formation process of placenta,thus speculate that CD82/KAI1 in embryo decidual process of feto-maternal conversation has a certain function.Objective: To study the role of CD82/KAI1 in the process of decidualization in mice on the D1-8.Methods: 1.The expression of CD82/KAI1 m RNA and protein was observed by the in-situ hybridization and immunofluorescence technique in D1-8 uterus of pregnant mice.2.To establish mouse uterine decidualization model in vivo and the expression of CD82/KAI1 protein was observed by immunofluorescence technology in the uterus on D5-D8 mice.3.After overexpressed or interfered with CD82/KAI1 expression,the protein expression of Cyclin D3,PR and CD82/KAI1 was detected by Western Blotting in the decidual cells.Results:(1)In situ hybridization and immunofluorescence assay showed that CD82/KAI1 was expressed in spatio-temporal specificity during the decidualization of mouse uterus.(2)In vivo,uterine decidua model is established.Immunofluorescence and Western blotting results also confirmed CD82/KAI1 expression,and the expression pattern was similar to that in normal pregnancy.(3)Endometrial stromal cells was induced in vitro,and the results of Western Blot showed that the expression of CD82 was increased with the decidualization process.Conclusion: CD82/KAI1 may participate in the process of regulating the orderly decidualization of endometrial stromal cells after embryo implantation.
