CD82/KAI1is a widespread tumor suppressor that can inhibit the activity and invasion of tumor cells. The tumor is one of the most serious hazards to human health which is critically characterized of invasion and metastasis, and this is also an important indicator of malignancy. It is found very similar to the invasive ability of trophoblast cells and the behavior of tumor cells in the mammalian embryo implantation.The trophoblast cells invade the maternal endometrium and form the placenta together with the decidua during the mice pregnancy in order to ensure normal gestation.The over-invasion or invasive deficiency in trophoblast cells would both cause harm to the mother and the embryo. Based on the preliminary experimental results,it was suggested that CD82expressed in the mouse endometrium which indicated that CD82/KAI1Gene could make an influence on decidual cells to promote the invasion of trophoblast cells.Objective:To study the expression of CD82/KAI1in the mouse uterine decidual cells and ectoplacental cone as well as its influence on the invasion of ectoplacental cone. Method:1. The well-designed CD82/KAI1interferential sequence was inserted into pSES-HUS vector, build pAdeasy-SES-HUS-CD82siRNA, and pAdeasy-SES-HUS carrier,2.The mouse uterine stromal cells were cultured and induced to decidualization in vitro, and the expression of CD82/KAI1were detected in decidual cells by RT-PCR and western blot.3.The D8mice uterus were made into paraffin sections which showed the expression of CD82/KAI1in extoplacental cone and decidua by immunohistochemical techniques;4. The growth of mouse ectoplacental cone was observed by the co-culture with decidua.Results:1.The interferential adenovirus were successfully reorganized with the titer of7.2×1011pfu/ml;2.The CD82/KAI1mRNA and protein were significantly expressed in mouse decidual cells;3.The CD82/KAI1protein were found in the mouse ectoplacental cone too by Immunofluorescence;4.The CD82/KAI1could inhibit the invasion of mouse ectoplacental cone to uterus endometrial.Conclusion:1.The CD82/KAI1were expressed in mouse uterine decidual cells and ectoplacental cone.2.The mouse uterine decidual cells express CD82/KAI1to inhibit the invasion of the ectoplacental cone.
