Objective: To investigate the expression of GODZ in temporal cortex of intractable TLE patients and PTZ-kindled epileptic rats and to discuss its possible role in the epileptogenesis.Methods: Temporal lobe cortex were collected from 14 intractable temporal lobe epilepsy patients and 20 traumatic brain injury patients.PTZ-kindled rats model were induced by intraperitoneal injection of pentylenetetrazol.The cellular distribution of GODZ in brain tissues of both TLE patients and rats was determined using double-labled immunofluorescence.And the level of GODZ protein and mRNA between TLE patients,pentylenetetrazole(PTZ)-kindled rats and controls was measured using immunohistochemistry,western blot and polymerase chain reaction.Results: GODZ expression was identified on cytomembranes and in cytoplasm of neurons in temporal neocortex of TLE patients,and it was also identified in hippocampal and cortical tissues of PTZ-kindled rats.Butin astrocytes,no GODZ expression was found.Decreased GODZ protein and mRNA were identified in brain tissues of TLE patients and PTZ-kindled rats compared to controls.Conclutions: GODZ is expressed in neurons but not astrocytes,and epilepsy may reduce the protein and mRNA level of GODZ,indicating a possible role of GODZ in the pathogenesis or pathophysiology of epilepsy. |
