In this study we investigated the regulation of TLR4 signaling in macrophages by zinc finger protein zbtb20. We found that silencing of zbtb20 could inhibit LPS-induced production of proinflammatory cytokines and interferon-beta in macrophages and accordingly suppress activation of MAPK (ERK, JNK and p38) pathway. The similar phenomenon was obserbed in LPS-stimualted macrophages derived from zbtb20-deficient mice. In contrast, zbtb20 overexpression promoted TLR4-triggered cytokine production and MAPK activation. Therefore, we demonstrate that zbtb20 may function as a positive regulatory molecule in TLR4 signaling.
