Objective:To observe the intervention effect of Shenqi Anti-tumor Decoction on liver cancer in Kunming mice,and to explore the mechanism of invasion and metastasis of Shenqi Anti-tumor Decoction side to liver cancer.Methods:Select 60 Kunming mice which weight 20±2g.The inoculated with H22hepatoma cells,the rats were randomLy divided into six groups,10 mice in each group,including model group,cisplatin group,Shenqi Anti-tumor Decoction high dose group,Shenqi Anti-tumor Decoction low dose group and Shenqi Anti-tumor Decoction high dose cisplatin combined with cisplatin group,Shenqi Anti-tumor Decoction suppressor low dose group.After second days of inoculation group to drug treatment,Shenqi Anti-tumor Decoction high and low dose groups were given 1g crude drug/m L,0.5g crude drug/mL,for 14consecutive days.Cisplatin group and cisplatin were injected into cisplatin in high and low doses of cisplatin group and Shenqi suppressor side,and the administration of cisplatin was0.05mg/0.2m L/d.After 14 days,the blood was extracted from the eyeball,and the tissues of liver cancer and spleen were taken.Observation index(1)the condition of mice;(2)Spleen index,tumor weight and tumor suppressor rate;(3)Changes of tumor histomorphology(4)The content of CD3~+,CD4~+and CD8~+in serum;(5)gene expression of MMP2,MMP9 and VEGF;(6)The protein expression of MMP2,MMP9 and VEGF.Result:(1)There was a significant difference in weight change between the treatment group and the model group(P<0.05).(2)The high and low dose combined cisplatin group were 63%,44%,36%and 23%in high and low dosage groups,and 52%in cisplatin group,respectively,and the high and low dose of cisplatin group were 63%and 44%respectively.(3)Compared with the model group,the spleen index of each treatment group increased to varying degrees.The high dose group of Chinese medicine and the combination of Chinese medicine and cisplatin increased significantly compared with the positive group,and the difference was statistically significant(P<0.05).(4)Compared with the model group,the concentration of CD8~+and CD3~+,CD4~+in all treatment groups increased,and the concentration of CD8~+and the concentration of CD3~+and CD4~+in the Chinese medicine group increased significantly compared with those in the cisplatin group(P<0.05).(5)Histopathology showed that the number of Chinese medicine group of hepatocellular carcinoma HCC cell necrosis is more unequal in size,lack of uniform nuclear staining;traditional Chinese medicine combined with cisplatin group tumor cell density decreased,cell degeneration necrosis increased,more connective tissue hyperplasia,liver cells showed lumps.(6)Compared with the model group,the expression levels of MMP2,MMP9,VEGFmRNA and their protein in cisplatin group,high dose Chinese medicine group and Chinese medicine combined with cisplatin group were all decreased,and the expression level of mRNA in traditional Chinese medicine combined with cisplatin group decreased significantly.Compared with the positive group,the difference was statistically significant(P<0.05).Conclusion:Shenqi Anti-tumor Decoction can inhibit the invasion and metastasis of H22 hepatoma cell xenografts and play a coordinated role with cisplatin.Its mechanism of action is related to improving the immunity of the body,reducing the related factors of tumor invasion and metastasis,and inhibiting the formation of tumor invasion and metastasis.
