Objective: To study the effects of Dendrobium nobile polysaccharides(DNP)on lifespan and its related indexes of C.elegans,and to explore preliminary its mechanism.Methods: DNP groups(0.06,0.12,0.24 mg/m L),PBS buffer and blank control group(0 mg/L)were established to investigate the effects of DNP on the lifespan and physiological indexes(reproductive capacity,pumping rate and body swing frequency)of N2 wild-type caenorhabditis elegans.To perform oxidative and heat stress test of elegans,and investigate preliminarily the mechanism of DNP retard aging.Results:(1)The ordinary lifespan test: compared to blank control group,the average lifespan of elegans in the DNP groups was prolonged significantly,and it was prolonged by 12.55%(0.06mg/ml group),15.19%(0.12mg/ml group)and 33.89%(0.24mg/ml group)(all P<0.05),while the average lifespan of PBS buffer group was not statistically significant different(P>0.05).(2)The reproductive capacity test: compared to blank control group,the reproductive capacity of each DNP groups was not statistically significant different(all P>0.05).(3)The pharynx-pumping rate and body-swing rate measurement: pharynxpumping rate and body-swing rate of each experimental group decreased with the increase of age,and compared with the blank control group,nematode pharyngeal pump and body swing frequency in the DNP groups were not statistically significant different(all P>0.05).(4)The oxidative stress test: compared to the blank control group,average lifespan of elegans in DNP groups was prolonged significantly,and it was prolonged respectively by 27.91%(0.06 mg/ml group),32.30%(0.12 mg/ml group)and 41.86%(0.24 mg/ml group)(all P<0.05),while the average lifespan in the PBS buffer group was not statistically significant different(P>0.05).(5)The heat stress test: compared to blank control group,average lifespan of elegans in DNP groups was prolonged significantly,and it was prolonged respectively by 16.27%(0.06 mg/ml group),18.05%(0.12 mg/ml group)and 19.08%(0.24 mg/ml group)(all P<0.05),while the average lifespan of the PBS buffer group was not statistically significant different(P >0.05).Conclusion: DNP can retard the aging process of caenorhabditis elegans,and the mechanism might involved in increase the stress resistance ability of elegans.
