The Extraction And Purification Of Dendrobium Nobile Lindl. Alkaloids And Its Effect On The Activity Of C.elegans

Dendrobium nobile Lindl.is a traditional Chinese medicinal material which was recorded in ancient books.With the in-depth study of dendrobium,it can not only be used as ornamental potted plant,but also can be used in medicine or health food.Dendrobium nobile Lindl.for asparagus,Orchidaceous,Dendrobium,rich in alkaloids,polysaccharides and phenolic substances compared with other congeneric Dendrobium,Dendrobium nobile Lindl.alkaloids content is relatively high,so this topic research the Dendrobium nobile Lindl.optimum conditions of extraction of total alkaloid and alkaloids purification methods,and explore the alkaloid extracts in Caenorhabditis elegans(C.elegans)on the activity of influence,the main research results of this paper is as follows:(1)Optimize the ultrasonic extraction method of total alkaloids from Dendrobium nobile Lindl.Use the aid-dye ofnrimetry to determine the content of alkaloids.Perform the orthogonal optimization based on four single-factor experiments:ultrasonic time,solid-liquid ratio,ultrasonic power and ethanol volume fraction.The results showed that ultrasonic power and ethanol volume fraction had significant effects on the total alkaloid extraction content of Dendrobium nobile Lindl.(P<0.05).The best extraction conditions were ultrasonic extraction time 25 min,solid-liquid ratio 1:16,ultrasonic extraction power 200 w,when the volume fraction of ethanol is 90%,the total alkaloids produced in Guizhou Dendrobium can be up to 5.08(mg/g).(2)The AB-8 macroporous resin was selected as the optimal material for purification by static adsorption and analytical experiments.The optimal conditions for the purification were as follows:loading volume 4 BV,80%alcohol elution concentration,and 1.0 mL/min flow rate pH 4.0,loading 0.3 g/mL,stem height ratio 1:9.The purified material was detected by HPLC with a content of 3.16 mg/g.The methodological investigation results showed that the precision,repeatability and stability were all good.The RSD was lower than 10%,the recovery rate was 99.14%,and the RSD value was 1.66%.(3)The purified alkaloids of Dendrobium nobile Lindl.were applied to C.elegans,and the effects of their ability to act on C.elegctns and the action of pharyngeal pumping were observed.The results showed that alkaloids had a positive effect on the C.elegans locomotor activity(P<0.05),and had no significant effect on the feeding behavior of C.elegans(P>0.05).The effects of alkaloids on the growth and development of C.elegans were further investigated and the results showed that they were effective against C.elegans.Both body length and body length have a positive effect(P<0.05);however,in the experiment on the reproductive ability of C.elegans,it was not found that it would affect the C.elegans's oviposition ability and oviposition cycle;In the experiment on the life-span of C.elegans,it was found that the average lifespan of C.elegans was prolonged(P<0.05),and the average survival time of worms in oxidative stress environment was significantly increased(P<0.05).
