Objective: To observe the effect of Dendrobium nobile polysaccharides oncells proliferation、insulin secretion and the mRNA expression of Ins,Pdx1and MafA byINS-1cells cultured in high glucose.Methods:1.We divided into six groups include normal control group (NC), high glucosecontrol group (HG), rosiglitazone control group (ROZ), Dendrobium nobilepolysaccharides low dose group (LD), middle dose group (MD), high dose group (HD),and each group was detected by CCK-8after48h’s foster.2. After48h’s foster, discardingthe old culture fluid in each group, while adding high-glucose medium were incubated1heach group, and extracted the supernatant fractions. Then measured the insulin levels bychemiluminescence enzyme immunoassay.3. The mRNA expression levels of Ins、Pdx1and MafA was detected by Real-time PCR.Results:1. Compared with NC group, HG group had lower rate of cell proliferation(P<0.05), but compared with HG group, MD and HD groups cell proliferation wassignificantly increased (P<0.05).2. Detected insulin secretion, HG group was lower thanNC group, while relative to HG group, MD and HD groups improved the insulin secretion(P<0.05).3. qRT-PCR results showed that, compared with NC group, Ins、Pdx1and MafAmRNA expression of HG group was reduced (P<0.05), while MD and HD groupsincreased the role of these three genes mRNA expression (P<0.05).4. In each of the aboveindicators, compared LD group with HG group, they had no significantdifference(P>0.05).Conclusion: In a long time under high glucose levels, Dendrobium nobilepolysaccharides can enhance INS-1cell proliferation, enhance the secretion of insulin,increase expression levels of Ins, Pdx1and MafA mRNA.
