Objectives To investigate the relationship between changes of IL-4 and IL-12 in vaginal microenvironment and high-risk human papillomavirus(HPV)infection and cervical lesions.Methods The patients who visited in the Department of Obstetrics and Gynecology,Affiliated Hospital of North China University of Technology in Tangshan City from January 2016 to January 2017 with cervical HR-HPV positive was selectde as research object.Colposcopic examination were conducted of positive cervical HPV result and colposcopic cervical biopsy were performed.According to histopathological results,176 cases were marked as subjects,among which 67 cases were HPV infected but histopathologically negative group.There were 63 cases with low grade intraepithelial lesions,33 cases with high grade intraepithelial lesions,and 13 cases with cervical cancer.Double-antibody sandwich enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of IL-4 and IL-12 in the vaginal microenvironment.Results 1 The levels of IL-4 in the vaginal microenvironment of HPV infection group,LSIL group,HSIL group and SCC group were 36.39±19.55,74.74±3.24,82.66±7.29,and 104.69±14.96 pg/ml,the difference was statistically significant(F=173.163,P<0.05),with the severity of cervical lesions,IL-4 levels gradually increased.2 The levels of IL-12 in the vaginal microenvironment of the HPV infection group,LSIL group,HSIL group,and SCC group were 90.56±1.23,89.42±0.36,89.08±0.48,and 88.85±0.05 pg/ml,the difference was statistically significant(F=37.612,P<0.05),with the severity of cervical lesions,IL-12 content gradually decreased.3 The ratios of IL-4/IL-12 in the vaginal microenvironment of the HPV infection group,LSIL group,HSIL group,and SCC group were 0.40±0.22,0.84±0.04,0.93±0.08,and 1.18±0.17,the difference was statistically significant(F=179.499,P<0.05),with the severity of cervical lesions increasing,the ratio of IL-4/IL-12 gradually increased.4 In the HPV infection group and the LSIL group,12 other high-risk HPV infections were predominant;in the HSIL group and SCC group,HPV16-type infections were predominant,and the difference was statistically significant(?~2=20.086,P<0.05).5 There was no significant difference about the levels of IL-4 in the vaginal microenvironment between HPV16,18 and 12 other high-risk HPV groups(P>0.05).There was no significant difference about the levels of IL-12 in the vaginal microenvironment between HPV16,18 and 12 other high-risk HPV groups(P>0.05).There was no significant difference about IL-4/IL-12 in the vaginal microenvironment between HPV16,18 and 12 other high-risk HPV groups(P>0.05).The level of IL-4 and IL-12 in the vaginal microenvironment has not been found to be related to the type of HPV infection.Conclusions 1 With the aggravation of cervical lesions,the content of IL-4 in the vaginal microenvironment increased,the content of IL-12 decreased,and the ratio of IL-4/IL-12 increased.2 In the HPV infection group and the LSIL group,12 other high-risk HPV infections were predominant;in the HSIL group and SCC group,HPV16-type infections were predominant.3 This study has not found that the different types of HPV infection are related to the changes of IL-4 and IL-12 levels in the vaginal microenvironment.
