Objectives To observe the effect of Kudiezi injection on renal ischemia-reperfusion injury in rats and explore its possible mechanism.Methods 32 male Sprasue-Dawley(SD)rats were randomly divided into sham operation group(Sham group),model group(IRI group),kudiezi injection group(KDZ group),the group of kudiezi injection was divided into low dose group(Low does group)and high dose group(High does group).The serum creatinine(Cr)and blood urea nitrogen(BUN)were detected by automatic biochemistry analyzer.The activity of superoxide dismutase(SOD)and malondialdehyde(MDA)in renal tissue were respectively assessed by detection kits.Histopathological changes were observed with HE staining.The expression of interleukin 6(IL-6)in renal tissue was checked by immunohistochemistry.The content of intercellular adhesion molecule-1(ICAM-1)was tested by ELISA.Results Compared with the Sham group,the content of Cr and BUN and MDA in renal tissue in the group IRI were significantly increased,and the content of Cr and BUN and MDA in renal tssue in group KDZ were between the Sham group and the IRI group,the above three indexes of low dose group were all higher than those of high dose group.In group IRI,the activity of SOD in kidney tissue was lower than that in group Sham,the activity of SOD in group KDZ was between Sham group and IRI group,and SOD activity in low dose group was lower than that in high dose group.HE stain showed that the morphology and structure of renal tubulars in group Sham were basically normal,Only mild edema in the epithelial cells,Compared with group Sham,there were vacuolar degeneration of renal tubular epithelial cells and expansion of part lumen in group IRI,and a large number of exfoliated cells and tubules were seen in them.Interstitial edema was obvious,and a large number of inflammatory cells were infiltrated.The renal tubular epithelial cells in group low dose were swollen than those in group Sham,the lumen narrowed,and the exfoliated epithelial cells in the lumen were less than those in group IRI,with slight edema in interstitial tissue,and a small amount of inflammatory cells infiltration.The degree of pathological damage was lighter than that in group IRI.In group High dose,the swelling degree of renal tubular epithelial cells was between Sham group and Low dose group.Epithelial cells shedding less,interstitial edema slightly,and minimal inflammatory cell infiltration.The degree of pathological damage was lighter than that in Low dose group.Immunohistochemical staining showed that the expression of IL-6 in the Sham group was slight,the expression of IL-6 in the IRI group was heigher than that in the Sham group,and the expression of IL-6 in the KDZ group was lower than that in IRI group,the expression of low dose group was higher than that in high dose group.ELISA detection show that the content of ICAM-1 in group IRI and group KDZ were all higher than that in the Sham group,but the content of ICAM-1 in the KDZ group was lower than that in IRI group,and the index in low dose group was higher than that in high dose group.Conclusions The KDZ injection can alleviate ischemia-reperfusion injury in rats,its mechanism is related to anti oxidative stress and reducing inflammatory injury.
