Objectives:By preparing a monkey model of ischemic stroke,observe whether experimental macaques show depressive symptoms,and provide evidence for early diagnosis,early prevention,early treatment and pathogenesis of PSD.Methods:Six adult normal male macaques aged 7-9 years and weighing 7-9 kg were randomly selected.Four MCAO rats were used to prepare middle cerebral artery ischemia(MCAO)models.Two of them were used as control groups.Success;SPECT was used to detect regional cerebral blood flow changes in experimental groups of macaques,and task-oriented neurological rating scale,post-embolus neurological rating scale,standard neurological rating scale and stroke clinical rating scale were used to evaluate the experimental monkeys Postoperative neurological function,close observation of postoperative neurological recovery of experimental monkeys to stable,and then comparing the experimental group of macaque postoperative weight changes;using focused video recording observation behavior of rhesus monkeys;macaques,sugar water test,determination of rhythm monkeys pleasure The lack of degree;comprehensive evaluation of experimental monkeys macaque post-stroke depression.Results:1.All four monkeys were modeled successfully,including 2 MCAOs on the right,2 MCAOs on the left,and MCAO macaques on the left because of vascular variability and massive cerebral infarction.They died on the second day after operation.2.In the experimental group,macaques had the most severe neurological deficit on the 1st day after surgery,and they were significantly injured compared with the preoperative ones(P<0.05).The neurological function of the macaques gradually recovered.There was no significant difference between the four months after surgery and preoperative(P>0.05).3.In the experimental group,the body weight of macaques was reduced to the lowest level at 14 days after surgery,and there was no significant difference compared with that before surgery(P>0.05).Subsequently,the weight of macaque monkeys gradually recovered.At the 4th month after surgery,the two experimental groups of macaque monkeys surpassed preoperatively,but there was no significant difference(P>0.05).4.The ratio of cerebral blood flow in the experimental group to the cerebral blood flow in the contralateral normal brain tissue reached the lowest at 14 days after operation,and there was no significant difference between the two groups at 3 days after operation(P>0.05).Cerebral blood flow gradually recovered.At 1 month after surgery,the ratio of cerebral blood flow on the infarct side to that on the contralateral normal brain tissue was significantly increased compared with 14 days after surgery.There was a significant difference between the two groups(P<0.05).5.In the experimental group,macaques were compared with the control group and the sugar water test was conducted in the half year after surgery.The percentage of preference of the macaques in the experimental group did not change significantly(P>0.05).Macaque monkeys did not show the core symptoms of depression anhedonia.6.Compared with the control group,monkeys in the experimental group were measured for activity within half a year after surgery.There was no significant change in the daytime activity of the macaques in the experimental group(P>0.05),but there was a significant decrease in activity at night(P<0.05)..7.In the experimental group,compared with the control group,interest and behavior were recorded in the first half of the operation.The interest and behavior of the monkeys in the experimental group were significantly reduced.The number of environmental exploration behaviors(P>0.05)and duration(P>0.05)was significantly shortened.Compared with the control group,monkeys in the experimental group had emotional and behavioral records for half a year after surgery.Macaque monkeys in the experimental group did not experience the depression of depression typical of the experimental group.The specific manifestations were the number of contractures(P>0.05)and duration(P>0.05).No significant increase occurred.Conclusions:1.Successful preparation of ischemic stroke macaque model with middle cerebral artery occlusion;2.In the experimental group,the weight loss,cerebral blood flow,and neurological impairment of the rhesus monkeys after the model were established,but the model macaques began to recover in one week after surgery,and returned to normal in the fourth month after operation;3.In the experimental group,half of the year after model establishment,except for the decrease in environmental exploration behavior,there was no depressive-like behavior such as depression,activity decrease,and anhedonia.Therefore,under the condition of single-cage feeding,the model monkeys do not suffer from post-stroke depressive symptoms that may be manifested in the community life of stroke patients.
