Objective:The expression level of NPR3 protein in patients with different types of osteosarcoma was detected by immunohistochemical method.By analyzing the relationship between the expression of NPR3 protein and the prognosis of osteosarcoma patients,it was discussed whether NPR3 protein could be used as a clinical marker to guide the malignant degree and prognosis of osteosarcoma.The molecular mechanism of NPR3 protein regulating osteosarcoma development and metastasis is preliminarily discussed.Methods:This subject collected 189 cases of osteosarcoma in Yunnan Tumor Hospital(Third Affiliated Hospital of Kunming Medical University)from November 2006 to March 2015,and divided into osteosarcoma group of osteoblast,fibroblastic osteosarcoma group,fibroblast osteosarcoma group and small cell osteosarcoma group according to its pathological types.The group is then grouped according to whether or not it is transferred.The differential expression of NPR3 protein in paraffin section samples of patients with osteosarcoma in different groups was detected by immunohistochemical method,and the difference between the 4 groups was analyzed The metastasis status was compared between the 4 groups,and long Rank was used to analyze the correlation between the 4 groups.SPSS 17.0 statistical software was used to analyze the results(P<0.05 showed statistical significance).Results:The 189 patients with osteosarcoma were divided into 4 groups according to their pathological types:osteoblast,fibroblast,fibroblast and small cell type.Immunohistochemical staining showed that all of the NPR3 proteins were expressed in the above cases and their location was in cytoplasm.The total number of metastatic osteosarcoma patients was 100 cases,of which 28 cases were highly expressed in NPR3 protein and 72 cases were low expression.The total number of osteosarcoma patients without metastasis was 89 cases,80 cases were highly expressed in NPR3 protein,and 9 cases were low expression.Statistical analysis showed that the expression level of NPR3 protein in the metastatic group was significantly different from that in the non metastatic group(P<0.01).There were 53 cases of osteoblast type and 26 cases in the metastatic group,of which 7 cases were high expression of NPR3,19 cases with low expression of NPR3,27 cases without transfer group,24 cases with high expression of NPR3 and 3 cases of low expression of NPR3.There were 48 cases of fibroblastic type and 21 cases in the metastatic group,of which 5 cases were high expression of NPR3,16 cases with low expression of NPR3,27 cases without transfer group,24 cases with high expression of NPR3 and 3 cases of low expression of NPR3.There were 44 cases of chondrocyte type and 18 cases in the metastasis group,of which 4 cases were high expression of NPR3 and 14 cases with low expression of NPR3;26 cases without metastasis group,23 cases with high expression of NPR3 and 3 cases of low expression of NPR3.There were 44 cases of small cell type and 35 cases in the metastasis group,of which there were 12 cases of high expression of NPR3,23 cases with low expression of NPR3,9 cases with no metastasis group,8 cases with high expression of NPR3 and 1 cases of low expression of NPR3.Statistical analysis showed that the difference between the above groups was statistically significant(P<0.01).The survival period of osteosarcoma in different groups showed that the median survival period of osteosarcoma was 33 months,the median survival period of fibroblastic osteosarcoma was 30 months,the median survival period of chondrosarcoma was 29 months,and the median survival period of small cell osteosarcoma was 21 months.Statistical analysis:the survival period of the small cell osteosarcoma group was significantly different from the osteosarcoma of osteoblast,chondroid osteosarcoma and fibroblast type osteosarcoma.There was no significant difference between the other three groups and 22 groups.Conclusions:The expression level of NPR3 protein may be an independent prognostic factor for the survival time of patients with osteosarcoma.
