OBJECTIVE:To investigate the expression of bone morphogenetic protein-2(BMP-2),osteopontin(OPN),38kD—protein kinase(P38)in osteosarcoma and its association with lung metastasis,and investigate the its association between them.METHODS:Using immunohistochemical method to detect the expression of BMP-2 and OPN and P38 in 46 osteosarcoma tissues which had complete follow up data.Result:1 ) BMP-2 and OPN expression was all located in the cytoplasm of oseteosarcoma cell,and P38 protein was observed in nucleus of osteosarcoma cell. The expression rate of BMP-2 in osteosarcoma was 58.70%(27/46).The expression rate of OPN was 71.74% (33/46). The positive expressionrate of P38 was 67.39% (31/46).The positive rates of BMP-2 andOPN and P38 in osteosarcoma tissue with lung metastasis were 83.33% (15/18),88. 89% (16/18) and 88.89% (16/18),respectively,which were much higher than that in osteosarcoma tissue without lung metastasis (BMP-2: 42.86%,12/28andOPN: 60.71%,17/28 and P38: 53.57%,15/28). BMP-2 and OPN and P38protein positive rates were not related to gender,age,size,location of tumor and osteosarcoma classification,but significanttly correlated with the clinical stage,differentiation degree and lung metastasis.2) There was a significantly positive correlation between the OPN and BMP-2 expression in osteosarcoma.And there was a significantly positive correlation between the P38 and BMP-2 expression in osteosarcoma.Conclusion:1) BMP-2,OPN and P38 is over expressed in osteosmcoma。2) The high expression of BMP-2 and OPN and P38 involve in the development of osteosarcoma and correlated closely to the lung metastasis of osteosarcoma..3) The expression of BMP-2 is closely correlated to that of OPN and P38 protein.
