Objective:Anti-Parkinson effect of Rosmarinic acid in vitro and in vitro and its possible mechanisms.Methods:1.C57BL/6 mice were randomly divided into high-dose rosmarinic acid group,low-dose rosmarinic acid group,positive group,model group,and control group,10 in each group.In the model group,MPTP(30mg/kg)was administered intraperitoneally to C57BL/6 mice for 5 days;the control group was given the same dose of 0.9%saline.The administration group was given the corresponding drug treatment while giving MPTP.The mice were continuously administered for 28 days,and the body weight changes of the mouse were observed every day and recorded.2.The open-field test used in the behavioral test was used to detect the autonomous activities of the mouse for each group 1,7,and 28 days after administration.The expression of TH and ?-synuclein in the brains of mice in each group were analyzed by western blotting at 1,7 and 28 days after administration.3.Take C57BL/6 pregnant mouse that have been pregnant for 18 days,dislocate the cervical spine,remove the fetus,and remove the fetal mouse brain tissue aseptically to separate and purify the neurons.The neurons were cultured in a 37 ?carbon dioxide incubator.After 24 hours,the neurons were cultured for half a week.After 72 hours,Ara-C(final concentration 2.5 mg/1)was used to purify the neurons.Cultivate until day 7 for experimentation.4.Through immunofluorescence experiments,the expression of TH and a-synuclein in the control group,MPTP group,and rosmarinic acid-administered group were observed by confocal microscopy.5.The neurons were treated differently and divided into control group,high-dose group of rosmarinic acid,middle-dose group of rosmarinic acid,and low-dose group of rosmarinic acid.The viability of neurons in each group was detected using the CCK-8 test kit;the effect of rosmarinic acid on the level of ROS in neurons was detected using a reactive oxygen test kit;Proteins associated with neuronal apoptosis were detected by western blotting:expression of caspase-3 and cleaved caspase-3 proteins.Results:1.Rosmarinic acid can improve the weight loss and the level of locomotor activity in a mouse model of Parkinson's disease;increase the level of TH protein in the brain of Parkinson's disease mouse model;and reduce the level of a-synuclein protein in the brain of Parkinson's disease mouse2.Rosmarinic acid can protect the MPTP-induced neuronal damage;inhibit the abnormal increase of a-synuclein;significantly reduce neuronal ROS levels;and prevent the abnormal activation of caspase-3.Conclusion:Rosmarinic acid can affect the pathological changes of Parkinson's disease mouse model and protect MPTP-induced neuronal damage.Its mechanism may be related to the inhibition of ROS release and the inhibition of apoptosis-associated protein caspase-3 activation.
