In this paper, The HPLC fingerprint system of Dendrobium huoshanense, Dendrobium officinale, Dendrobium moniliforme from huoshan was established through optimization of extraction and chromatographic conditions. Then, the fingerprint differences in stems, leaves and flowers between three Dendrobium species were compared.(1) Establishment of the optimal HPLC chromatographic fingerprint method.For compound extraction, methanol was the best suitable extractant and the optimal extraction time was 2 hours. For HPLC, under the detection wavelength of 270 nm, acetonitrile (A) and 0.1% acetic acid (B) as effective mobile phase was used to gradiently elute as follows:30% A (10 min),30-35%A (10 min),35-55% A (20 min), 55-70%A(5 min),70-100% A(25min),100%A(20 min)(2) Comparison of the fingerprints of stems, leaves, flowers between three Dendrobium species.The seven peaks whose retention times were at 8.12±0.09 min,10.58±0.11 min, 13.83±0.25 min,16.55±0.22 min,33.85±0.08 min,36.68±0.15 min and 72.62±0.13 min, respectively, displayed mainly differences in stems between three Dendrobium species and could be used in discrimination of three Dendrobium species. There were 10 characteristic peaks in leaves fingerprints, which were richer than that in stems. The peak at 8.24±0.14 min combined with other 5 peaks at 16.50±0.08 min,19.67±0.12 min,32.04±0.15 min,33.33±0.08min and 61.63±0.20 min could be used to identify the leaves of three Dendrobium species except in July, August and September. There were distinguished peaks at 31.70±0.25 min in Dendrobium moniliforme flowers and 74.29±0.12 min in Dendrobium huoshanense flowers. The peaks at 56.57±0.02 min could be detected in flowers of Dendrobium huoshanense and Dendrobium moniliforme, but it can not be detected in Dendrobium officnale.(3) Determination of naringenin, erianin and total phenols of three Dendrobium species.A similar variation in naringenin and erianin was found by comparing the 2nd peak and the 4th peak to the standards and both peaks began to increase and reached the highest point in the end of summer, then, quicky reduced to the lowest point during autumn to winter. It was also found that the august was crucial season for the accumulation of small molecules in Dendrobium, in which the content of naringenin, erianin and total phenols reached the highest levels in stems of three Dendrobium species.
