Dendrobium,which is one of valuable Chinese traditional medicine,firstly was set out in the Eastern Han Chinese“Shen Nong Ben Cao Jing”.It have been widely used in the treatment of weakness after febrile diseases,thirsty,fire excess from yin deficiency,asthenia-syndrome and other diseases.Modern pharmacological experiments have proved that they have a variety of active anti-tumor,immunomodulatory,anti-oxidation,lowering blood glucose and other activities.Dendrobium is surged in demand and becoming a natural green products enjoys a high reputation both at home and abroad.However,the shoddy phenomenon is more and more serious because of the shortage of valuable Dendrobium.Traditional Chinese medicine fingerprint technology is recognized as the most effective method of traditional Chinese medicine quality control at home and abroad.This study choose different origin of Dendrobium moniliforme,huoshanense and officinale as the research object,using the traditional Chinese medicine fingerprint technology,comparativing study the three kinds of Dendrobium of high performance liquid chromatography(HPLC)characteristic spectrum,to provide methods for its quality control level,in order to make the authenticity,advantages and disadvantages,effectively control the quality of Dendrobium.Objective1.To establish HPLC characteristic spectrum analysis method of different origin of Dendrobium moniliforme,to provide the basic methods for quality control and identification.2?To preliminarily compare Dendrobium moniliforme of Anhui with Dendrobium huoshanense by HPLC characteristic spectrum.3?To preliminarily compare Dendrobium moniliforme of Zhejiang with Dendrobium officinale by HPLC characteristic spectrum.Methods1?Use the HPLC characteristic spectrum analysis method of Dendrobium moniliforme to analysis different origin of samples,specific chromatogram similarity evaluation system software writed by the State Pharmacopoeia Commission Specific chromatogram similarity evaluation system software(2004A version)was adopted to generate a total of mode,comparing different origin of Dendrobium moniliforme commonalities and differences,and the similarity evaluation.2?Compare Dendrobium moniliforme of Anhui with Dendrobium huoshanense under the characteristic spectrum and similarity evaluation.3.Compare Dendrobium moniliforme of Zhejiang with Dendrobium officinale of Zhejiang under the characteristic spectrum and similarity evaluation.Results1.The establishment of Dendrobium moniliforme of different origin HPLC characteristic spectrum analysis method is feasible and practical.(1)Dendrobium moniliforme of Anhui(yellow Dendrobium moniliforme)were marked out of a total of 24 common peaks,which peak 15 and Peak 16 were their exclusive features common peaks,and the batches of samples' similarty were 0.821?0.995;Dendrobium moniliforme of Yunnan(plue Dendrobium moniliforme)were marked out of a total of 27 common peaks and the batches of samples'similarty were 0.869?0.937;Dendrobium moniliforme of Zhejiang(plue Dendrobium moniliforme)were marked out of a total of 22 common peaks and the batches of samples' similarty were 0.907?0.942;Dendrobium moniliforme of Zhejiang(yellow Dendrobium moniliforme)were marked out of a total of 28 common peaks and the batches of samples' similarty were 0.972?0.973.(2)The characteristic spectrum of Dendrobium moniliforme of Anhui(yellow Dendrobium moniliforme)to be a mutual mode control,the similarity of Dendrobium moniliforme of Zhejiang(yellow Dendrobium moniliforme)were 0.378,indicating that similarity of HPLC characteristic spectrum of Dendrobium moniliforme of Anhui and Dendrobium moniliforme of Zhejiang were very low and the difference was significant.(3)The characteristic spectrum of Dendrobium moniliforme of Anhui(yellow Dendrobium moniliforme)to be a mutual mode control,the similarity of Dendrobium moniliforme of Zhejiang(plue Dendrobium moniliforme)and Dendrobium moniliforme of Yunnan(plue Dendrobium moniliforme)were respectively 0.426 and 0.490,indicating that similarity of them were very low and the difference was significant.(4)The characteristic spectrum of Dendrobium moniliforme of Yunnan(plue Dendrobium moniliforme)to be a mutual mode control,the similarity of Dendrobium moniliforme of Zhejiang(plue Dendrobium moniliforme)were 0.960,indicating that similarity of HPLC characteristic spectrum of them were very high and the difference between different resources of plue Dendrobium moniliforme was not significant.(5)The biggest difference of the characteristic spectrum of Dendrobium moniliforme of Anhui was in the retention time of 80?120 min with significantly large proporti'on of 2 absorption peak group.2n The HPLC characteristic spectrum analysis method Dendrobium moniliforme of Anhui(yellow Dendrobium moniliforme)and Dendrobium huoshanense were established.(1)Dendrobium moniliforme of Anhui(yellow Dendrobium moniliforme)were marked out of a total of 24 common peaks and the batches of samples' similarty were 0.821?0.995;Dendrobium huoshanense were marked out of a total of 40 common peaks and the batches of samples' similarty were 0.959?0.988.(2)The characteristic spectrum of Dendrobium moniliforme of Anhui(yellow Dendrobium moniliforme)to be a mutual mode control,the similarity of Dendrobium huoshanense were 0.492,indicating that similarity of HPLC characteristic spectrum of them were low and the difference was significant.(3)Combination with the kinds of Dendrobium in the retention period the characteristic peaks of spectrum UV analysis.In the retention time of 80?120 min,the biggest difference of the characteristic spectrum of Dendrobium moniliforme of Anhui(yellow Dendrobium moniliforme)and Dendrobium huoshanense were the number of characteristic peaks,peak shape and the peak ratio.And Dendrobium moniliforme of Anhui(yellow Dendrobium moniliforme)have some typical characteristic peaks of flavonoids,and Dendrobium huoshanense' s peak area is not large but more specific set of characteristics.(4)The biggest difference of the characteristic spectrum of Dendrobium moniliforme of Anhui(yellow Dendrobium moniliforme)was in the retention time of 80?120 min with significantly large proportion of 2 absorption peak group,which could be identificated of Dendrobium huoshanense.3?The HPLC characteristic spectrum analysis method Dendrobium moniliforme of Zhejiang(yellow Dendrobium moniliforme)and Dendrobium officinale of Zhejiang were established.(1)Dendrobium moniliforme of Zhejiang(yellow Dendrobium moniliforme)were marked out of a total of 28 common peaks and the batches of samples'similarty were 0.972?0.973;Dendrobium officinale of Zhejiang were marked out of a total of 31 common peaks and the batches of samples' similarty were0.920?0.937.(2)The characteristic spectrum of Dendrobium moniliforme of Zhejiang(yellow Dendrobium moniliforme)to be a mutual mode control,the similarity of Dendrobium officinale of Zhejiang were 0.775,indicating that they had differences.Conclusion1?The analysis method of HPLC characteristic spectrum of Dendrobium moniliforme accurate and reliable,has a good stability,provided the methodology basis for quality control for Dendrobium.2.As the Dendrobium moniliforme of different origin are different in HPLC characteristic spectrum,this method can be used to preliminarily identify resources of Dendrobium moniliforme.3?This method can effectively distinguish Dendrobium moniliforme of Anhui and Dendrobium huoshanense,when Dendrobium huoshanense are posed with Dendrobium moniliforme of Anhui in Anhui market,which is one value of this study.4?This method can effectively distinguish Dendrobium moniliforme ofZhejiang and Dendrobium officinale of Zhejiang,when Dendrobium officinale of Zhejiang are posed with Dendrobium moniliforme of Zhejiang in Jiangzhe market,which is another value of this study.
