Research BackgroundHuman chorionic gonadotropin(HCG)is a pregnancy-specific marker that can be used to determine pregnancy by measuring its concentration.Currently,urine HCG concentrations are generally measured if qualitative tests are performed.If the quantitative tests are performed,the blood samples need to be collected.And the current quantitative detection method still has some shortcoming,such as long detection time,complex detection steps and so on.At present,the rate of acute myocardial infarction(AMI)keeps increasing.Studies have shown that cardiac troponin I(cTnI)is a highly specific and sensitive marker of myocardial damage.If AMI patient has chest pain,cTnI serum concentration will increase rapidly in a short time and has a dynamic change.According to the change of cTnI serum concentration and evidence of myocardial ischemia,doctor can determine if a patient is AMI.So it is very important that rapid and quantitative detection of cTnI serum concentration.However,the methods of cTnI detection also have some shortcoming,such as long detection time,complex detection steps and so on.Immunochromatographic assay(ICA)is a rapid diagnostic method that combines chromatography and immunolabeling.ICA not only has the characteristics of antigen-antibody reaction,but also takes membrane as reaction platform.ICA has some advantage,such as simple operation,simple device,real-time and reliable result and so on.ICA can be used to detect many markers.Superparamagnetic nanoparticles(SPMNPs)have good dispersibility and maneuverability.It can be coupled with a variety of biologically activegroups.Research purpose and significanceThe study aimed to establish magnetic Immunochromatographic test strips for rapid,quantitative detection of HCG,cTnI.The two kinds of test strips not only can provide accurate result,but also overcomes the shortcoming of long detection tome and complex detection steps.It is good for rapid,quantitative detection out of the hospital.Research content and methodThis study consists of two parts experiment:HCG experiment and cTnI experiment.In both experiment,the corresponding immune magnetic beads is made by EDC/NHS method and characterized by some methods.Then,we set up the test strips and verify the immune magnetic beads.Research results1:coupling rate of HCG immune magnetic bead:45ug anti-a-HCG-antibody/lmg SPMNPs;2:the limitation of the qualitative detection for HCG:1mIU/ml;the limitation of the quantitative detection for HCG:0.27mIU/ml;3:the type of NC membrane of HCG magnetic Immunochromatographic test strips:Sartorius CN95;4:coupling rate of cTnI immune magnetic bead:15ug anti-cTnl-antibody(16A11)/1mg SPMNPs;5:the limitation of the qualitative detection for cTnI:1ng/ml;the limitation of the quantitative detection for cTnI:0.46ng/ml6:the type of NC membrane of cTnI magnetic Immunochromatographic test strips:Millipore 135;7:both experimental results suggest that the antigen concentration is proportion to the color depth of T line.Research conclusionIn this study,two kinds of immune magnetic beads were used as probe to establish two kinds of magnetic Immunochromatographic test strips for rapid detection.The two kinds of strips can provide both qualitative and quantitative detection.
