Cardiac troponin ??cTn??,which was only found in cardiac tissue,was highly specific for myocardial injury and was used as the gold standard for the diagnosis of acute myocardial infarction?AMI?.It is of great significance for early diagnosis,risk stratification,and prognosis of AMI.Previously,our group first reported an innovative full-quantitative system by combining the magnetic nanoparticles with classic lateral flow chromatography assay,which named magnetic quantitative immunochromatography?MICA?.In this study,first,we optimized input amount or input ratio of key materials to prepared a noval Au@Fe3O4 flower-like magnetic nanoparticles with advantages of high-loaded and low background,and then they were applied in MICA to developed a rapid,low-cost,accurate and more highly sensitive cTn? detection method.Coupling buffer,antibody input amount,blocking agent and chromatography working solution were optimized seperately.Especially,great attention has been paid to the antibody types?including 5 of anti-cTn? specific monoclonal antibodies,1 of Tnc antibody and 2 of cTn complex?and combinations?17 combinations in total?,and a lot of research work has been done.Finally,the proposed method had a linear range of 0.075–20ng/m L with the limit of detection of 0.0149 ng/m L.It had good repeatability and all detection can be completed in 15 minutes.While detecting cTn? in 48 clinical serum,the detection results of target method were coincident with those by Beckman Coulter Access 2.Therefore,our method is expected to become a new auxiliary approach for the early diagnosis and prognosis of AMI,and there would be a great application potential in Emergency,community clinic,chest pain center and other special scenes.
