Objective: In this study,we try to explore whether acupuncture can regulate the expression of miRNA-124 in the striatum of transgenic mice with Parkinson's disease;to explore whether acupuncture can inhibit the aggregation of ?-synuclein protein in striatum by regulating the expression of miRNA-124,so as to improve the symptoms of transgenic mice with Parkinson's disease;and to explore whether acupuncture affects the expression of p-CREB,BDNF protein by regulating the expression of miRNA-124,and participates in the pathogenesis of Parkinson's disease through miRNA-124/CREB/BDNF signaling pathway.Methods: By Transforming SNCA*A53T gene into C57BL/6J mouse to establish PD model mouse,we divided them into four groups: negative control group,model group,acupuncture group and antagonist group.Acupuncture group was treated with acupuncture on the basis of the model group,and the antagonist group was treated with acupuncture and miRNA-124 antagonist on the basis of the model group.We adopted electroacupuncture technique to deal with acupuncture group and antagonist group.The parameters of electroacupuncture instrument were continuous wave,2 Hz for the frequency,1 m A for the intensity,lasted 30 minutes,once a day for 2 weeks.We selected "Yanglingquan" and "Taichong" for acupuncture points.The behaviour of these mice in different experimental groups were tested before and after acupuncture treatment by suspension test and swimming test.The expression of miRNA-124 in the striatum of transgenic mice with Parkinson's disease was detected by using RT-q PCR.The expression of ?-synuclein,p-CREB and BDNF protein were detected by Western blot.Results:(1)The score of suspension test and swimming test: Before acupuncture intervention,the negative control group was significantly higher than that in model group,acupuncture group and antagonist group,and the difference among them were statistically significant(P< 0.05),while there were no difference among model group,acupuncture group and antagonist group(P> 0.05).After acupuncture intervention,the acupuncture group and antagonist group were significantly improved compared with those before acupuncture intervention(P< 0.05),but there was no significant difference between acupuncture group and antagonist group(P> 0.05).(2)The expression of miRNA-124 in the striatum of mice: Compared with the negative control group,the expression of miRNA-124 in model group was significantly lower(P< 0.05).While compared with the model group,the expression of miRNA-124 in the striatum of acupuncture group increased significantly(P< 0.05),which could be blocked by the miRNA-124 antagonist.(3)The expression of ?-synuclein in the striatum of mice: The model group was significantly higher than that of the negative control group(P< 0.05),while the acupuncture group and antagonist group were down-regulated compared with the model group(P< 0.05),but the down-regulation of ?-synuclein expression in acupuncture group was more significant than that in antagonist group(P< 0.05).(4)The expression of p-CREB and BDNF in the striatum of mice: Compared with the negative control group,the expression of p-CREB and BDNF in the striatum of the model group was down-regulated(P< 0.05),but they can be reversed by acupuncture treatment(P< 0.05).But there was no significant difference between antagonist group and model group(P> 0.05).Coclusions:(1)Acupuncture may reduce the aggregation of ?-synuclein protein in striatum by up-regulating the expression of miRNA-124,thus improve the behavior of transgenic mice with Parkinson's disease,but its mechanism may be multiple.(2)Acupuncture can up-regulate the expression of p-CREB and BDNF protein,miRNA-124/CREB/BDNF signaling pathway may be involved in the pathogenesis of Parkinson's disease.
