Objective:To investigate serum adropin concentrations in type 2 diabetes?T2DM?patients,especially those overweight/obese,evaluate the relationships between adropin levels and metabolic parameters and explore the correlation of serum adropin with T2 DM and obesity.Methods:A total of 116 T2 DM patients and 60 normal controls?NC?were recruited to the study.Body mass index?BMI?and waist to hip ratio?WHR?were calculated.Each group was then divided into two subgroups including overweight/obesity and normal weight based on BMI?25 or 18.5-25 kg/m2.Anthropometric characteristics were collected and biochemistry including triglycerides?TG?,low-density lipoprotein cholesterol?LDL-C?,high-density lipoprotein cholesterol?HDL-C?,total cholesterol?TC?,fasting plasma glucose?FPG?,high-sensitive C reactive protein?hs-CRP?,glycosylated hemoglobin A1c?HbA1c?and fasting insulin?FIns?were detected by clinical laboratory.Using HOMA2 Calculator software,the subject's homeostasis model insulin resistance index?HOMA2-IR?was calculated based on the concentration of FIns and FPG at the same time.Adropin concentration was determined using commercial ELISA kits.Data analyses were performed using the SPSS version 21.0.Results:?1?Serum adropin levels in T2 DM patients were decreased compared with the normal control[3.77?2.96-5.49?vs.5.53?3.75-7.89?ng/mL,P<0.01].Meanwhile,overweight/obese patients had more considerably reduced level of adropin.?2?Serum adropin level of overweight/obesity group was lower than that of normal weight group within study participants.?3?Adropin level was negatively correlated with BMI,WHR,hs-CRP,TG,FPG,HbA1c,FIns,HOMA2-IR?r=-0.346,-0.392,-0.230,-0.270,-0.349,-0.359,-0.502,-0.513,respectively,P<0.01?,while positively with HDL-C?r=0.313,P<0.01?in study participants.The correlations of adropin with WHR and glucolipid variables?TG,HDL-C,FPG,FIns,HOMA2-IR,HbA1c?were still unchanged after adjusting the effect of BMI.Multiple linear stepwise regression analysis indicated that HOMA2-IR,WHR and HbA1c were independent influencing factors of serum adropin concentration??=-1.109,-6.836,-0.139,respectively,P<0.05?.?4?Binary logistic regression analysis showed that low adropin levels might be a risk factor for T2 DM even after adjusting for demographic or anthropometric variables and lipid profile?P<0.01?.?5?Receiver operator curve?ROC?demonstrated the optimal cut-off value of adropin concentration for confirming T2 DM was 4.07 ng/mL with sensitivity of 56.0%and specificity of 73.3%.Conclusion:Serum adropin concentrations are decreased in T2 DM patients.Meanwhile,serum adropin level of overweight/obesity group is lower than that of normal weight group.Low adropin concentrations are associated with an increased risk of metabolic diseases such as T2 DM and obesity,which may exert potential metabolic effects through modulation of energy balance,insulin resistance,lipid anabolism,and inflammatory responses.
