The Effect Of Echinops Laizfolius And It’s Compound Aqueous Extract On Glucose And Lipid Metabolism Of Type Ⅱ Diabetes Mellitus Rat Model

In order to develop a kind of new Chinese herbal which are able to prevent and control type Ⅱ diabetes mellitus, we explored the effect of Echinops laizfolius as well as it’s compound aqueous extract on glucose and lipid metabolism of type Ⅱ diabetes mellitus rat model. Firstly, type Ⅱ diabetes mellitus rat models were established by means of feeding high calorie and high sugar diet to male SD rats, intraperitoneal injection with streptozotocin (STZ) simultaneously. Secondly, rat models were randomly divided into control group, model group, Echinops laizfolius water extract of high dosage group (2.0g/mL), Echinops laizfolius water extract of middle dosage group (1.0g/mL), Echinops laizfolius water extract of low dosage group (0.5g/mL), Echinops laizfolius compound water extract of high dosage group (2.0g/mL), Echinops laizfolius compound water extract of middle dosage group (1.0g/mL), Echinops laizfolius compound water extract of low dosage group (0.5g/mL), Metformin group and Xiaoke pill group, different drugs were given by gavage for 56 days. During the period, the weight, glucose and lipid metabolism level, histological section of liver and pancreas were detected. The results are as follows:On the basis of having developed type Ⅱ diabetes mellitus rat models successfully, with the methods of feeding high calorie and high sugar diet to male SD rats, intraperitoneal injection with streptozotocin (STZ) simultaneously, there have no obvious effect of Echinops laizfolius as well as it’s compound water extract on weight, fasting blood-glucose, hepatic glycogen, muscle glycogen of type Ⅱ diabetes mellitus rat models (P>0.05); Glucose tolerance AUC of Echinops laizfolius compound water extract of high dosage group was reduced evidently compared with model group (P<0.05); Echinops laizfolius compound water extract group with high, middle and low dosage could reduce the content of TC,TG and LDL-C significantly(P<0.05), but the HDL-C had no significant difference (P>0.05); Serum insulin level and IRI (Insulin Resistance Index) were reduced in Echinops laizfolius water extract of middle dosage group and Echinops laizfolius compound water extract of high dosage group (P<0.05), there was no statistical difference between ISI (Insulin-sensitivity Index) (P>0.05).Pathology observation of livers:Except Echinops laizfolius water extract of low group, light or moderate fatty degeneration were found in liver cells of rats in the rest of the medicated groups. Pathology observation of pancreas:Pancreas islet was enlarged, with a distinct define from exocrine gland in Echinops laizfolius compound water extract of middle dosage group.Conclusion:Type II diabetes mellitus rat models could be developing successfully by the method of feeding high calorie and high sugar diet to male SD rats, intraperitoneal injection with streptozotocin (STZ) simultaneously.Echinops laizfolius as well as it’s compound aqueous extract could maintain the weight of type II diabetes mellitus rat models and improve Impaired glucose tolerance effectively.Echinops laizfolius as well as it’s compound aqueous extract could improve metabolic abnormality of lipid.Echinops laizfolius as well as it’s compound aqueous extract could reduce insulin resistanceEchinops laizfolius as well as it’s compound aqueous extract could repair the hobbled liver and pancreas of type II diabetes mellitus rat models.
