Osteosarcoma is one of the most common malignant tumor in the orthopaedic research.For its rapid development,easy to transfer,poor prognosis and other characteristics,it has always been valued by clinicians.The pathogenesis of osteosarcoma is extremely complex.which involves the interaction of multiple genes.It is the result of gene regulation network imbalance that causes abnormal metabolism of cells.Gene expression can usually be regulated from transcriptional level and posttranscriptional level.MicroRNA,lncRNA and circRNA,these three non-coding RNAs play very important roles in gene expression regulation.Moreover,these three non-coding RNAs and mRNA also have a wide range of association and cooperative regulatory relations,and several kinds of RNA are composed of numerous regulatory networks.Through the overall analysis of the abnormal molecular function regulation network in osteosarcoma,it can provide a great help for us to define the molecular mechanism of the occurrence and development of osteosarcoma.MiRNA,lncRNA and circRNA can regulate gene expression by ce RNA mechanism.More and more studies have shown that mi RNA,lnc RNA and circRNA play an indispensable role in the occurrence,development and metastasis of tumor.To study the complex molecular metabolic regulatory network composed of functional molecular interactions,such as mRNA and functional proteins,we analyzed systematically the specific non-coding RNA expression profiles of osteosarcoma tissues.It will not only help to explain the molecular mechanisms of osteosarcoma in the early stages of cancer.It can also provide a theoretical basis for early warning of osteosarcoma,design of targeted therapy drugs and clinical individualized treatment.It is expected to bring significant economic and social benefits.This study used the whole transcriptome sequencing and bioinformatics technologies to construct the regulation network of non-coding RNA in osteosarcoma.And analyzed the target genes regulated by non-coding RNA in the network.By analyzing the differential noncoding RNA expression profiles.3 pairs of osteosarcoma tissues and their adjacent normal tissues,we screened 163 differentially expressed lncRNA in osteosarcoma tissue,of these,67 were up-regulated and 96 were down-regulated.We screened 156 differentially expressed circRNA,of these,64 were up-regulated and 92 were down-regulated.And We screened 205 differentially expressed miRNA,of these,90 were up-regulated and 115 were down-regulated.In addition,the molecular regulation network based respectively on lncRNAs and circRNAs as the core was constructed by using DIANA-mirPath version 3.0 and RegRNA 2.0.Use R language to perform functional annotation analysis respectively on lncRNAs and circ RNAs molecular regulation network,Inc RNAs and circRNAs regulatory network were found to regulate the cell proliferation,differentiation,apoptosis and other biological processes in the development of osteosarcoma.And it is closely related to the various metabolic pathways involved in regulation of actin cytoskeleton,polypeptide processing and tumor metabolism.
