Part 1 Expression and significance of serum TNF-α,IL-1β,MCP-1and HYP in patients with rare earth pneumoconiosis objective The contents of TNF-α,IL-1β,MCP-1and HYP in serum of patients with rare earth pneumoconiosis were measured,and the sensitive indexes of early auxiliary diagnosis of rare earth pneumoconiosis were discussed.Methods 26 patients diagnosed with rare earth pneumoconiosis in a city in recent three years were selected as rare earth pneumoconiosis group,and 47 people who took part in physical examination at the same time were selected as control group.Fasting venous blood was taken at 5 ml.The expression of TNF-α,IL-1β and MCP-1,HYP in serum was detected by enzyme-linked immunosorbent assay(Elisa).Results The serum levels of IL-1β and HYP in patients with rare earth pneumoconiosis were higher than those in the control group,while the levels of TNF-α and MCP-1 in the patients with rare earth pneumoconiosis were lower than those in the control group(P<0.05).In rare earth pneumoconiosis group,the expression level of IL-1β and MCP-1 increased gradually with the increase of age(P<0.05).The expression level of IL-1β in serum of smoking group was higher than that of non-smoking group(P<0.05),but there was no significant difference among the other three indexes,and the expression level of HYP increased gradually with the increase of rare earth pneumoconiosis stage(P<0.05).The expression of IL-1β and MCP-1,HYP in serum of patients with length of service <12 years was significantly lower than that of patients with12 years of service(P<0.05).The expression level of HYP was positively correlated with length of service after excluding age,smoking and age(P<0.05).Conclusion The differential expression of TNF-α,IL-1β and MCP-1,HYP in serum of rare earth pneumoconiosis patients is of great significance for the development of rare earth pneumoconiosis.IL-1β and HYP can be used as sensitive indexes for early diagnosis of rare earth pneumoconiosis.Part 2 Comprehensive Analysis of Aberrantly Expressed Profiles of LncRNAs and MiRNAs with Associated ceRNA Network in the Plasma of Patients With Rare Earth Pneumoconiosiss objective By exploring the differential expression profiles of miRNA,LncRNA,and mRNA in the plasma of patients with rare earth pneumoconiosis,a miRNA-LncRNA-mRNA ceRNA regulatory network was constructed to provide an experimental basis for further studying the pathogenesis of rare earth pneumoconiosis.Methods Nine people from the rare earth pneumoconiosis group and the control group were selected for plasma analysis and sequencing analysis of miRNA,LncRNA,and mRNA;differentially expressed miRNA,LncRNA,and mRNA were analyzed using GO and KEGG analysis;Use q-RT-PCR for verification;Use STRING database to construct protein interaction networks of differential genes;Use the Onco Lnc online platform to analyze the top five mRNAs obtained from PPI network analysis,and differentially expressed miRNAs and lncRNAs in lung cancer Study for survival analysis.Results We found 125 differentially expressed LncRNAs(up-regulated 49,down-regulated 76),5differentially-expressed miRNAs(all down-regulated),and 82 differentially-expressed mRNAs(up-regulated 38,down-regulated 44).The GO analysis found that the items mainly enriched in differentially expressed lncRNAs were the response of cells to extracellular stimuli,the negative regulation of smooth muscle cell proliferation,and Polo-like kinase activity;the items that were mainly enriched in differentially expressed miRNAs were the regulation of GTPase activity,DNA repair,ion migration,and so on;the main enriched items of differentially expressed mRNAs are transmembrane signalling receptor activity,autophagic vacuole formation,splicing of nuclear mRNA by splicing,and cellular response to extracellular stimuli.By KEGG analysis,differentially expressed lncRNAs are mainly enriched in the mutual conversion of pentose and glucuronide,fructose and mannose metabolism,spliceosome,etc.;differentially expressed miRNAs are mainly enriched in Hedgehog signaling pathway,c AMP signaling pathway,glycan Lipid metabolism,pantothenic acid,and Co A biosynthesis;differentially expressed mRNAs are mainly concentrated in autophagy regulation,Notch signaling pathway,and hematopoietic cell lineage.We constructed an lncRNA-miRNA-mRNA ceRNA regulatory network.Q-RT-PCR showed the same trend for sequencing results of partially differentially expressed RNA,and sequencing results were reliable.In lung squamous cell carcinoma,the expression levels of hsa-mi R-16-2-3p and KRR1 are positively correlated with the overall survival rate of patients,P < 0.05;in lung adenocarcinoma,the expression level of CEP83-AS1 is positively correlated with the overall survival rate of patients Correlation,the expression level of SNW1 was negatively correlated with the overall survival rate of patients,P < 0.05.We also found that 56 of the 125 differentially expressed lncRNAs were distributed in the testes.Conclusion A total of 125 lncRNAs,4 miRNAs,and 82 mRNAs were differentially expressed in the plasma of patients with rare earth pneumoconiosis.Differential expression of RNA is mainly related to the response of cells to stimulation;the main pathways of enrichment are Notch signaling pathway and autophagy regulatory pathway.The construction of the lncRNA-miRNA-mRNA ceRNA regulatory network in rare earth pneumoconiosis revealed the interactions among various key lncRNA,miRNA,and mRNA in the occurrence and development of rare earth pneumoconiosis.Rare earth dust can cause damage to workers’ reproductive systems. |
