Mesenchymal stem cells?MSCs?are first found in bone marrow,and are named for their ability to differentiate a variety of mesenchymal like phenotypes.MSC,as a member of the stem cell family,which is derived from early mesoderm,is a pluripotent stem cell with significant self renewing and multiple differentiation potential.Mesenchymal stem cells could be obtained from the umbilical cord,placenta,bone,fat and connective tissue,and under the specific environment and cytokines,could also be induced and differentiate into many tissue cells,such as myocardial cells,fat cells,nerve cells,liver cells and osteoblasts.Recent studies also show that the extrom,containing different RNA and protein complexes,secreted by mesenchymal stem cells is also important in the growth and differentiation of multiple cells.In addition,mesenchymal stem cells have less immunogenicity and no severe immune rejection during allograft.It is an ideal source of cell repair and remodeling for clinical treatment,and has broad application prospects.Liver is the largest essential organ of the body and plays an important physiological function in the body.Liver injury is the basis of liver failure.There are many reasons for liver injury,including viral infection,toxic substances intake,improper medication and hepatic ischemia reperfusion.Acute liver injury may appear in a short period of time,such as abnormal liver function,jaundice and other clinical manifestations.Severe or persistent damage can eventually lead to cirrhosis and even liver failure.Carbon tetrachloride?CCl4?is the most commonly used chemical injury model for induced tube,which can induce acute liver injury in a short time,and it can also produce chronic liver injury and liver cirrhosis model under repeated administration.The specific mechanism of carbon tetrachloride induced liver injury is not clear yet.However,it is generally believed that acute liver injury is a direct invasion of hepatocytes,destruction of enzyme protein synthesis and severe necrosis and lesions in the liver when CCl4 enters the body for a short time.Chronic liver injury is caused by CCl4 in vivo,which causes liver degeneration,necrosis and fibrous tissue hyperplasia caused by long-term and repeated injury,and then cirrhosis.In the construction of liver injury models,there are many differences in the dosage,time and route of specific dosages reported in different literatures.However,closed group mice or SD rats are usually used for hepatotoxicity test.In particular,C57BL/6mice are widely used for the construction of liver damage models because of their genetic stability and the consistency of phenotype.In this study,C57BL/6 mice were induced by intraperitoneal injection of carbon tetrachloride to construct both acute and chronic liver injury models.After comparing the two sets of models,we selected the appropriate model to enter human umbilical cord derived mesenchymal stem cells into the caudal vein and detected their therapeutic effects on chronic liver injury models.This study preliminarily confirmed that human placenta derived mesenchymal stem cells have certain therapeutic effect on chronic liver injury induced by carbon tetrachloride,and laid a foundation for mesenchymal stem cells from basic research to clinical application.
