Objective:To explore the relationship between NLRP3 gene polymorphism and hypertension in Xinjiang Kazakh.Methods:the First Affiliated Hospital of Xinjiang Medical University from February 2016 to March 2017 was selected randomly 230 cases of Xinjiang Kazakh primary hypertension and 220 cases of healthy Kazak people(Healthy control)were collected at heart center.Clinical and laboratory data were collected,and peripheral venous blood was collected.The genotype and allele of 2 SNPs loci(rs10754558,rs3806268)of NLRP3 gene were detected by Tapman probe method,and the distribution of genotypes and alleles in two groups was analyzed.The content of plasma interleukin 1(IL-1)beta was detected by ELISA.SPSS 18.0 software system are used to deal with the data.All the measured data are represented by mean standard deviation,and the count data are expressed in frequency.The comparison between Hardy-Weinberg equilibrium test,the gene frequency of four X~2 test table;the blood pressure and the index difference between different genotypes using two independent samples t test,P<0.05difference was statistically significant.Multivariate Logistic regression analysis was used to evaluate the correlation between various factors and hypertension.The difference between bilateral P<0.05 was statistically significant.Results:there was a significant difference in genotype distribution between rs10754558 and control group in hypertension group.The frequency of GG vs(GC+CC)gene in the hypertension group was significantly higher than that in the control group(P=0.01).After correcting the confounding factors with Logistic,the risk of rs10754558 genotype GG in hypertension group was significantly higher than that in control group(OR=2.436,95%CI:1.238-4.792,P=0.01).There was no significant difference in genotype distribution between rs3806268and control group in hypertension group,and there was no significant difference between hypertension group and control group in each group(genotype:P>0.05,allele:P>0.05).Compared with the control group,IL-1?group significantly increased plasma beta in hypertension[(2.79+0.83)ng/L and(2.21+0.91)ng/L,P<0.05],The plasma IL-1?levels in patients with hypertension with GG genotype were significantly higher than GC and CC genotypes[(34.05.7)ng/L VS(25.17.4),(24.19.0)ng/L,P<0.05].Conclusion:NLRP3rs10754558gene polymorphism and genetic susceptibility to essential hypertension related.There is no correlation between the polymorphism of NLRP3rs3806268 gene and the occurrence of Kazak hypertension.
