| Objective 1.To observe the effects of Danhuang Mingmu Decoction on SOD and MDA of BN rats after DR laser operation.2.To explore the effect of Danhuang Mingmu Decoction on oxidative stress in BN rats after DR Laser operation.3.To observe the effect of Danhuang Mingmu Decoction on the expression of retinal occludin and ZO-l protein in BN rats after DR Laser irradiation,and to explore the effect on blood-retinal barrier.Methods The BN rats were randomly divided into 4 groups.Group A was blank group,a total of 6 rats,and general feed was fed.Group B,C Group,D Group were model group,Chinese Medicine Group,Andomin group,8 in each group,all high fat feed feeding.The model of BN rats was induced by intraperitoneal injection of 40mg·Kg-1 STZ.When the blood glucose of BN rats reached 16.7mmol/L,the DM model was established.Begin to calculate the course of disease.After 8 weeks,each group of Group B,C and D randomly selected 2 rats to make HE stain to confirm the successful establishment of the DR model.532 argon laser photocoagulation was applied to the retinal photocoagulation of BN rats,observed one week,and then the FFA was used to observe the laser spot,which was a successful model for diabetic retinopathy after laser operation.The rats with blood sugar levels were treated as failure,and the blood sugar was restored to normal after the model was modeled.Group A and group B to saline irrigation stomach,group C to Chinese herbal compound granules 16ml/kg(equivalent to adult equivalent dosage)irrigation stomach,group D to andomin suspension 16ml/kg(equivalent to adult equivalent doses)irrigation stomach.are once daily,continuous irrigation of the stomach for 4 weeks.After 4 week(the 13 th week)s,the FFA was measured,the blood glucose,body weight,serum SOD,MDA index of BN rats were tested,and the expression of occludin and ZO-1 protein was detected by parallel he staining and Western-blot.Results 1.Blood sugar,weight: There was no significant difference in blood glucose levels in each group(P>0.05).After modeling,the blood glucose in Group A and B,C and D was significantly different in the course of 3 days,9 weeks and 13 weeks(P<0.01).There was no significant difference in weight before the model was made(P>0.05).During the course of 9 and 13 weeks,the weight of rats in Group A was significantly different than that in the corresponding B,C and D group(P<0.01).2.Serum SOD index: The activity of sod in Group C was significantly higher than the Group B,close to the Group A,there was no statistically significant difference between groups(=1.153,p>0.05),paired T test showed no statistical difference between Group A and Group B,C and D(p>0.05).3.Serum MDA index: Serum MDA level in Group B was higher than Group A,there were statistically significant differences between groups(=9.232,P<0.05).The difference between Group A and C,Group B and D wasn’t statistically significant(P>0.05),while Group C had statistically significant differences with Group B(P<0.05).4.Western-Blot detection: occludin protein relative expression,Group A,D compared with Group B,the difference wasn’t statistically significant(P>0.05);Group C and B comparison,the difference was statistically significant(P<0.01);ZO-l protein relative expression,Group D compared with Group B,the difference wasn’t statistically significant(P>0.05);Group A,C compared with Group B,the difference was statistically significant(P<0.01).Conclusion 1.BN rats with DR after Laser operation glucose continued to be high and weight continued to decrease.2.Danhuang Mingmu Decoction were not significantly improvement to reduce the blood sugar level and increase the weight of rats.3.Danhuang Mingmu Decoction can inhibit the oxidative stress level of rats’ retina.In this study,the content of MDA decreased and SOD content increased,which suggested that Danhuang Mingmu Decoction could improve the oxidative stress state of rat retina,remove free radicals and protect retinal cells from oxidative damage effectively.4.Danhuang Mingmu Decoction can increase the expression of occludin and ZO-1 protein in DR rat’s retina,in which the effect of Danhuang Mingmu Decoction is better.It is suggested that Danhuang Mingmu Decoction may be related to increasing the relative expression of occludin and ZO-1,thus affecting TJ’s bonding performance and repairing BRB damage.5.This experiment evaluated the effect of Danhuang Mingmu Decoction on the treatment of diabetic retinopathy after laser operation through the determination of oxidative stress and the expression of retinal tissue protein.It is suggested that Danhuang Mingmu Decoction can improve blood vessel permeability by inhibiting oxidative stress in the retina of rats,enhancing blood vessel resistance and repair BRB damage and improve retinal function. |
