Objectives To investigate the effect of Zhixue Mingmu recipe on inflammatory factors,oxidative stress index and advanced glycation end products in diabetic retinopathy rats,so as to determine whether it has therapeutic effect.Methods A total of 85 male SD rats were randomly divided into two groups,10 in normal group and 75 in experimental group,and 75 rats in the experimental group were subjected to a one-time intraperitoneal injection of 1% streptozotocin(45mg/kg)to induce a diabetic model.After successful induction of the diabetic model,the diabetic retinopathy(DR)rat model was confirmed to be successful by fundus fluorescein angiography.Subsequently,60 DR rats were randomly divided into model group,positive medicine(hydroxyl benzene sulfonic acid calcium capsules)group,Zhixue Mingmu recipe groups(low-dose group,medium-dose group,and high-dose group),12 mice per group.All rats were given gavage for 30 days in accordance with the experimental design.At the two and four week,the rats were weighed and their blood glucose was monitored by collecting tail vein blood.After the last administration,random blood glucose levels were detected in all rats.On day 30,Bilateral eyeballs were collected for retinal histopathological examination.The permeability of blood-retinal barrier was measured by Evans blue assay(EB).VEGFA,b FGF,CRP,s ICAM-1,IL-6,GSH-Px,AGEs and Ang-Ⅱ factors were detected by ELISA(enzyme linked immunosorbrent assay,ELISA).Results 1 Body weight and blood glucose: After two weeks of intragastric administration,the difference between the model group and the blank group was statistically significant(P< 0.05);There was no significant difference between the positive drug group,Zhixue Mingmu Recipe groups and the model group(P > 0.05).2 The results of HE staining showed that the structure of each layer of retina in blank group was clear,the cells were arranged regularly,and the morphology was not abnormal.Compared with the blank group,the retinal of model group was thinner and the ganglion cell layer showed vacuolar changes.The retinal cells in the positive drug group were thinner than the blank group but thicker than the model group,the cell arrangement was denser than the blank group,and the number of retinal ganglion cells was less.Compared with the blank group,the retinas in the control group were thinner and the number of dense retinal ganglion cells in the control group was decreased.The retinal thickness of medium-dose group was thicker than that of model group and positive drug group and the number of retinal ganglion cells was increased.The retinas of rats in the low-dose group were thinner than those in the blank group and had a lower number of dense retinal ganglion cells than those in the positive drug group.Compared with the model group,the retinal thickness in the medium-dose group was obviously thickened,with regular and compact cell arrangement.3 EB assay:The amount of EB leakage in the retinal tissue of rats in the model group was significantly higher than that in the blank group(P < 0.05);The amount of EB leakage in retinal tissue of positive drug group and Zhixue Mingmu Recipe groups was lower than that in the model group(P < 0.05).There was no significant difference in the amount of EB leakage between the medium-dose group and the high-dose group and the positive drug group.4ELISA results: VEGF-A,b FGF,CRP,SICAM-1,IL-6,AGEs and Ang-II factors in retinal tissue of model group were increased compared with blank group(P < 0.05).Compared with the model group,the expression of these factors in the retinal tissue of the positive drug group and the high-dose group decreased(P < 0.05).There was no significant difference between the high-dose group and the positive drug group(P > 0.05).GSH-Px in retinal tissue of the model group was lower than that of the blank group(P < 0.05).GSHPx in retinal tissue of positive drug group and high-dose group were increased compared with model group(P < 0.05).There was no significant difference between the high-dose group and the positive drug group(P > 0.05).Conclusions 1 Zhixue Mingmu formula has no effect on lowering blood sugar and affecting body weight.2 Zhixue Mingmu formula has a certain therapeutic effects on the early stage of diabetic retinopathy in rats by regulating inflammation,oxidative stress indexes and advanced glycation end products.Figure 13;Table 5;Reference 108... |
