Objective:Analysis after cystic resection syndrome patients and healthy people and patients with asymptomatic gut microbes after cystic resection basically colony information whether there is a difference,to explore may result in the pathogenesis of syndrome after cystic resection.Methods:1.CTAB method was used to treat 8 cases of patients with cholecystectomy syndrome,8 healthy people and 8.The genomic DNA of fresh fecal microorganism was extracted after cholecystectomy.2.choose cells ribosomes small subunits of 16 s rRNA genes on the High variation area,using specific primers with Barcode,New England Biolabs company High-Fidelity PCR Master Mix with GC Buffer and efficient hi-fi enzymes for PCR amplification.3.product recycling use of PCR amplification TruSeq(?)PCR-Free DNA Sample Preparation Kit for building a Kit for library construction,and use HiSeq2500 sequence PE250 computer.4.OTU clustering and species annotation,sample complexity analysisand multi-sample comparative analysis were performed on the data of sequencing.Results:1.24 cases feces samples to target the genomic DNA region PCR amplification of the OUT data clustering and species result has carried on the comprehensive statistics,can be used to build OTUs and obtain the number of Tags annotation information more than 97%.2.Intestinal waste microbial flora,relative abundance,in turn,is in the top 10 flora:Firmicutes,Bacteroidetes,Proteobacteria),Verrucomicrobia),Actinobacteria,Cyanobacteria,Tenericutes,Planctomycetes,Acidobacteria,Gemmatimonadetes.3.Groups of patients with post-cholecystectomy syndrome,group of healthy persons and patients with no symptoms after cholecystectomy had higher similarity of community structure in different groups.4.Compared with the healthy group,after cystic resection group of patients with syndrome of gut microbes mainly colony Verrucomicrobia,Bacteroidetes and Firmicutes information difference had no statistical significance(P>0.05);The difference of bacterial colony information of Proteobacteria was statistically significant(P = 0.0100).5.Compared with group of patients with asymptomatic after cholecystectomy,cholecystectomy syndrome after gut microbes in the patient group main colony Verrucomicrobia,Bacteroidetes,Firmicutes and Proteobacteria abundance of flora value difference has no statistical significance(P>0.05).Conclusion:1.Compared with a group of patients with asymptomatic post-cholecystectomy syndrome gut microbes in the patient group main colony abundance value differences without statistical significance.2.Compared with the healthy group,after cystic resection syndrome in patients with intestinal microorganism bacterium content is higher3.The increase of deformable bacteria in feces of patients with postoperative cholecystectomy may be the cause of chronic diarrhea and abdominal pain in patients with postcholecystectomy syndrome.
