| Objective:To investigate the protective effect of gastrodin on myocardial ischemia reperfusion injury in rats with myocardial ischemia reperfusion injury.Methods:The first part was randomly divided into sham operation group,I/R group and gastrodin treatment group(50mg/Kg,100mg/Kg,200mg/Kg).The myocardial ischemia/reperfusion injury model was induced by refilling 2h after the left anterior descending branch(LAD)of the rat heart.TTC staining was used to detect the area of myocardial infarction and to detect cardiac function in rats with langenddorff.The second part is divided into sham operation group,I/R group;Gastrodin pretreatment group,chloroquine group,gastrodin + chloroquine group,gastrodin + rapamycin group.Using Western blotting and immunohistochemical detection of autophagy protein LC3-? edge area of infarction,P62,Beclin 1 and LAMP-2 and apoptosis related proteins the Bcl-2 and the expression of Bax.Results:Compared with the I/R group,gastrodia pretreatment can reduce the area of myocardial infarction and protect myocardial function,with the best effect of 100mg/Kg(P<0.05).Western Blotting showed:gastrodia elata pretreatment group LC3 ?,P62,the expression of Beclin 1 also lower(P<0.05),while the expression of LAMP-2 elevated(P<0.05),and partly eliminate the effect of chloroquine;Compared with I/R group,bcl-2 protein expression was up-regulated(P<0.05),while Bax expression was down-regulated(P<0.05).The same results were also demonstrated in immunohistochemistry.Conclusion:It can improve the systolic and diastolic function of myocardial infarction after myocardial ischemia reperfusion injury in rats,and reduce the area of myocardial infarction after I/R injury.The protective effects of gastrodia on myocardial ischemia reperfusion injury in rats were preliminarily confirmed,which may be associated with the reduction of autophagy and the improvement of autophagy,and the reduction of apoptosis. |
