| Objective:The model of zebrafishes with microvascular deletion was established to st udy the effect of gastrodin on angiogenesis.The pharmacological effects of gast rodin on cerebral ischemia reperfusion injury were evaluated by the model of rats with middle cerebral artery occlusion reperfusion.The effect of gastrodin on neurogenesis was preliminarily studied.Methods:1.The vascular areas and the number of vascular branches under the intest ines were observed to evaluate the effects of gastrodin on angiogenesis.2.Middle cerebral artery occlusion model in rats was established by using the suture method.The model was evaluated by using neural deficit scores and TTC staining.3.The pharmacological effects of gastrodin on cerebral ischemia reperfusion injury were evaluated by using neural deficit scores,TTC staining,neurobehavioral tests,including novel object recognition,Morris water maze.4.Immunofluorescence was used to observe the effects of gastrodin on vW F and GFAP expression in MCAO rats.5.Western blot was used to study the expression of NeuN and GFAP.Results:1.Compared with the control group zebrafishes,the vascular areas and the number of vascular branches under the intestines in model group zebrafishes wer e reduced obviously.Compared with the model group zebrafishes,the vascular areas and the number of vascular branches under the intestines of zebrafishes in gastrodin were increased.2.The infarcted areas were showed on every brain tissue section of rats in model group.And the infarcted area is a third of the area of the whole brain section.The injury of neurological function in model group rats was serious.The neural deficit scores of MCAO rats in gastrodin group were lower than model group rats(P<0.01).Compared with the model group rats,the recognition ability of MCAO rats to novel objects was improved by treated with gastrodin.Morris water maze training found that the latency period(P<0.01)and the escape distances(P<0.01)of the model group rats were significantly longer than sham group rats.The time spent in the target quardant and the crossing times were reduced than sham group rats(P<0.01).Compared with the rats in the model group,the latency period and the escape distances of MCAO rats in gastrodin group were reduced in the Morris water maze training(P<0.01).The rats in gastrodin group got a larger number of crossing times and spent more time in the target quardant when they compared with the rats in the model group.3.The brains of rats in the model group were atrophied obviously.Compare d with the model group,the volume of cerebral atrophy of rats in the high dose(P<0.01)and low dose(P<0.05)of gastrodia significantly reduced.4.Compared with the sham group rats,the expression of vWF positive cells was increased in the model group rats.Compared with the model group rats,the expression of vWF were increased(P<0.01)in gastrodin group rats.Compared with the sham group rats,a large number of GFAP positive cells were showed in the cortex and striatum of rats in the model group.Compared with the model gro up,the high dose(P<0.01)and low dose(P<0.05)of gastrodin could reduce the expression of GFAP.5.Compared with the sham group,the expression of NeuN in the model group rats was decreased(P<0.05).Compared with the model group,the express ion of NeuN in the high dose and low dose of gastrodin group rats were significa ntly increased(P<0.05).Compared with the sham group rats,the expression of GFAP was increased in the model group(P<0.01),the expression of GFAP was reduced in the high dose and low dose of gastrodin group rats(P<0.05).Conclusion:1.Gastrodin can significantly promotes the angiogenesis of zebrafishes with microvascular delection.2.Gastrodine can obviously improve the neurological function of MCAO rats cased by cerebral ischemia reperfusion injury.Gastrodine can improves the learning and memory abilities of MCAO rats.Gastrodine can protect the brains by reducing cerebral atrophy.Gastrodine has significant effect on the prevention and treatment of cerebral ischemia reperfusion injury.3.Gastrodin can improve the expression of vWF in brain tissues of MCAO rats.It shows that gastrodin has obvious effect on promoting angiogenesis.Me anwhile,gastrodin can reduce the damages of brain tissues by decreasing rea ctive astrocytes.4.A preliminary study on the effect of gastrodin on regulating neurogenesis which caused by cerebral ischemia reperfusion injury.It is shows that gastrodin has the effect on promoting the differentiation of neural stem cells to mature neur ons. |
