Objective Osteosarcoma is the most common primary malignant bone tumor in adolescents,and it is resistant to the common chemotherapeutic drug cisplatin.This article investigated the effect of mi R-422 a on cisplatin's inhibition of osteosarcoma MG-63 cell proliferation and induction of cell apoptosis.Methods 1.The osteosarcoma MG-63 cells were transfected with mi R-422 a mimics.The experiments were divided into four groups: blank control group,mi R-422 a group,cisplatin group,and cisplatin+mi R-422 a group,The efficiency of transfection was observed under a fluorescence microscope,and osteosarcoma cells with high expression of mi R-422 a were established.2.MTT assay detected the proliferation rate of osteosarcoma MG-63 cells after high expression of mi R-422 a.3.Flow cytometry assay was used to detect the apoptosis rate of osteosarcoma MG-63 cells after mi R-422 a overexpression.Results 1.QPCR results showed that mi R-422 a transfected,its expression level was significantly higher than the blank control group(P<0.01).2.MTT assay results showed that the proliferation rate of osteosarcoma MG-63 cells was lower in the mi R-422 a group,the cisplatin group,and the cisplatin+mi R-422 a group than in the blank control group(P<0.05);In the cisplatin + mi R-422 a group,there was no significant difference between the cisplatin group and the mi R-422 a group(P<0.05);between the mi R-422 a group and the cisplatin group(P> 0.05).3.Flow cytometry assay results show:the apoptosis rate(%)of the blank control group,mi R-422 a group,cisplatin group,and cisplatin+mi R-422 a group cells were 5.69±0.79,14.44±1.53,13.89±1.92,and 23.73±2.85,respectively;the apoptosis rate of osteosarcoma MG-63 cells in mi R-422 a group,cisplatin group and cisplatin+mi R-422 a group was higher than that in blank control group(P<0.05);Cisplatin+mi R-422 a group was higher than mi R-422 a group and cisplatin group(P<0.05);mi R-422 a group was higher than blank control group(P<0.05);the cisplatin group was higher than the blank control group(P<0.05);the apoptosis rate of the mi R-422 a group was not significantly different from that of the cisplatin group(P>0.05).Conclusion mi R-422 a can increase the inhibitory effect of cisplatin on proliferation of osteosarcoma cell MG-63 and induce apoptosis.
