Adiponectin Inhibits Hepatocytic Lipoapoptosis By Up-regulating ACSL1

Objective: To confirm that Adiponectin inhibits hepatocytic lipoapoptosis by up-regulating Long-chain acyl-Co A synthetase 1(ACSL1),and provide a theoretical basis for the treatment of Adiponectin in the non-alcoholic fatty liver disease(NAFLD).Methods: Human L02 cells were cultured in vitro,and induced apoptosis of L02 cells by palmitic acid.Then they were divided Into 6 groups: A: control group,B: Adiponectin group C:Adiponectin+pc DNA3.1-NC group,D: Adiponectin+Si RNA-NC group,E: Adiponectin+Si RNA-ACSL1 group F: adiponectin pc DNA3.1-ACSL1 group.After transfection for 48 hours,Used western blot to detect the expression of ACSL1 protein in L02 cells.Used The biochemical kit to detect the content of triglyceride(TG)in human L02 cells.Used flow cytometry to detect the apoptosis of human L02 cells.Results: 1.The expression of ACSL1 protein in L02 cells in each group: group B compared with group A,the expression of ACSL1 protein inecreased(P < 0.01).group F compared with group B,the expression of ACSL1 protein inecreased(P < 0.01).group E compared with group B,the expression of ACSL1 protein decreased(P < 0.01).2.TG content of L02 cells in each group: group B,group C,group D,group E,group F compared with group A,the TG content decreased,Group F > Group B,Group C,Group D > Group E(P < 0.01).group C and group D compared with group B,the TG content is not significant difference(P>0.05).group F compared with group B,the TG content decreased(P < 0.01).group E compared group B,the TG content inecreased(P < 0.01).3.The total apoptosis rate of L02 cells in each group: group B,group C,group D,group E,group F,compared with group A,the total apoptosis rate decreased,Group F > Group B,Group C,Group D > Group E.(P < 0.01).group C and group D compared with group B,the total apoptosis rate is not significant difference(P>0.05).group F compared with group B and,the total apoptosis rate decreased(P < 0.01).group E compared with group B,the total apoptosis rate inecreased(P < 0.01).Conclusion: Adiponectin can inhibit the deposition of TG and lipid apoptosis of hepatocytes by up-regulating the expression of ACSL1 gene.
