Background and Objective In clinical cohort studies,the high expression of long-chain acyl-coenzyme A synthetases 1(ACSL1 gene)in peripheral white blood cells of patients with acute myocardial infarction(AMI)has been found to be utilized as molecular markers of myocardial infarction diagnosis.The plasma triglyceride level of the AMI patients is significantly higher than that of healthy people.We hypothesized that the high expression of ACSL1 gene increases the level of triglyceride,which is one of the pathogenesis of AMI promoted by ACSL1 gene.In this report,cytological method was adopted to clarify the hypothesis and further study the effect and mechanism of ACSL1 gene on lipid metabolism.Methods In this research,liver cells of healthy people were cultured,the overexpression and the interference vectors of ACSL1 were constructed and transfected into liver cells instantaneously.The content of intracellular triglyceride was quantitatively analyzed through Elisa method.the mechanism of the effect of ACSL1 gene on lipid metabolism was studied by PCR array technique.Results After overexpression of ACSL1,the m RNA expression of ACSL1 in the overexpression group was significantly higher than that in the negative control group(17.49 ± 5.10 vs 1.00 ± 0.05);The expression of ACSL1 protein was significantly increased,so were the triglyceride content(97.27 ± 2.22 vs 72.35 ± 0.44,P < 0.001).The expression level of fatty acid oxidation pathway PPAR? was significantly down-regulated compared with the control group(0.48 ± 0.05 vs 1.01 ± 0.20,P=0.041),So were the genes associated with fatty acid synthesis pathways: SREBP1,ACC,FAS and SCD1.After interfering with ACSL1,the m RNA expression of ACSL1 in the interferential group was significantly lower than that in the negative control group(0.54 ± 0.03 vs 1.00 ± 0.03,P <0.001);The expression of ACSL1 protein decreased,so were the content of triglyceride(130.65 ± 3.64 vs 142.07 ± 4.23,P=0.024).PPAR? was up-regulated(1.44 ± 0.22 vs 1.04 ± 0.34,P=0.158),while SREBP1,ACC,FAS and SCD1 were down-regulated compared with the control group.Conclusion In summary,ACSL1 genes affected triglyceride levels.High expression of the ACSL1 gene reduced fatty acid ?-oxidation through the PPAR? pathway,thereby increasing triglyceride levels.
