Objective:The experiment was designed to survey the expression of SOCS-3and SHP-1 in JAK-STAT signaling pathway of cancer tissues and its corresponding paracancerous tissues in HBV-related hepatocellular carcinomaand(HCC),and explore the relationship between their expression and clinical pathological parameters.Method:A total of forty-one patients with HBV-related HCC were collected from the first affiliated hospital of university of South China from 2012 to2017.The expression of SOCS-3 and SHP-1 was detected by immunohistochemical streptavidin-perosidase menthod.What's more,the relationship between the two factors expression and the patient's age,sex,liver function,alpha-fetoprotein(AFP),tumor diameter,HbeAg,hepatic cirrhosis,lymph nodemetastasis,the degree of differentiation,clinical staging and other clinical data were analyzed by?~2 Test.Result:1?The positive expression rate of SOCS-3 protein in HCC tissues andparacanceroustissueswere29.3%(12/41)and75.6%(31/41).Similarly,the positive expression rate of SHP-1 protein in HCC tissueswas24.4%(10/41),inparacanceroustissueswas70.7%(29/41)(P<0.05).It showed that SOCS-3 and SHP-1 protein were highly expressed in paracancerous tissues(P<0.05).2?In HBV-related HCC,the positive expression rate of SOCS-3 in middle-low differentiation was 12%(3/25)significantly lower than that in high differentiation 56.3%(9/16)(P<0.05).The positive expression rate of SHP-1 in middle-low differentiation was 8%(2/25)lower than that in high differentiation 50%(8/16)(P<0.05).The positive expression rate of SOCS-3 protein in hepatocellular carcinoma patients with cirrhosis was22.9%(8/35)lower than that in hepatocellular carcinoma patients without cirrhosis 66.7%(4/6)(P<0.05).The positive expression rate of SHP-1protein in hepatocellular carcinoma with cirrhosis was 14.3%(5/35)lower than that in hepatocellular carcinoma without cirrhosis 83.3%(5/6)(P<0.05).The rate of positive expression of SOCS-3 protein in A+B stage patients was 24.3%(9/37)in Barcelona Clinic Liver Cancer(BCLC)stage,which was lower than that in C+D stage 75%(3/4)(P<0.05).The positive expression rate of SHP-1 protein in A+B stage was 18.9%(7/37),which was also lower than that in C+D stage 75%(3/4)(P<0.05).The expression of SOCS-3 and SHP-1 protein in the experimental specimens were independent of the age,gender,AFP,tumor diameter,HBeAg,and antiviral therapy(P>0.05).Conclusion:1.In HBV-related HCC,the positive expression rate of SOCS-3 and SHP-1 protein in HCC tissues were lower than that in paracancer tissues.2.The expression of SOCS-3 and SHP-1 proteins in HBV-related HCC were connected with the degree of differentiation,cirrhosis and BCLC stage.
