Objective Wilms' tumor, a common solid malignant tumor in children, the molecular mechanism of its occurrence and development are not yet clear. The survival rate has been greatly improved by the current combined therapy, and how to improve the survival rate is always of the concerns of medical workers. Currently, the researches about diagnosis and treatment are much more than those about its occurrence, development and prognosis. The suppressor of cytokine signalings are kinds of negative regulation factors which generate by cell factors and can block cytokine signal transduction, by inhibiting the JAK/ STAT (Janus kinase/ signal transducer and activator of transcription) pathway to inhibit signal transductions, which SOCS-1 and SOCS-3 have the strongest actions.In this article we focus on investigating the expression levels of suppressor of cytokine signaling-1 and suppressor of cytokine signaling-3 in the Wilms' tumor.The pathological classifications have an important connections with the prognosis after operations and the survival rates,so we make the futher researches to study the inherent relationship between the expression levels of mRNA and the different pathological classifications, in order to provide the new method of gene therapy in Wilms1 tumor.Methods Collecting thirty Wilms' tumor tissue samples from the surgergs operated in Pediatric Surgery, provincial Hospital Of Shandong. Collecting ten puerile normal kidney tissues samples from the patients suffering from injuries and severe hydronephrosis.(All the samples were agreed by the family members of patients.)The expression levels of SOCS-1 and SOCS-3 were detected by RT-PCR and Western-Blot,and the dates were analysised by SPSS software.Results 1,The expression levels of SOCS-1 mRNA and SOCS-3mRNA: Compared to the normal kidney tissues,the expressions of SOCS-1 mRNA and SOCS-3mRNA in nephroblastoma were significantly lower,which having the statistically significants(F:18.253,21.174, P<0.01). The expressions of SOCS-1 mRNA and SOCS-3mRNA in adjacent tissues were lower than those in normal kidney tissues(F:6.031,7.105, P<0.05).2,The expression levels of protein of SOCS-1 and SOCS-3:Compared to the normal kidney tissues,the expressions of protein of SOCS-1 and SOCS-3 in nephroblastoma were significantly lower,which having the statistically significants(F:20.024,16.149, P<0.01). The expressions of protein of SOCS-1 and SOCS-3 in adjacent tissues were lower than those in normal kidney tissues(F:5.627, 6.202, P<0.05).3, The expression levels of SOCS-1 mRNA and SOCS-3mRNA in the different pathological classifications:The expressions of SOCS-1 mRNA and SOCS-3mRNA in UH were significantly lower than those in FH(F:4.974,5.596, P<0.05).Conclusion 1,The expression of mRNA and protein of the SOCS-1 and SOCS-3 in nephroblastoma were significantly lower which may illustrate SOCS-1 and SOCS-3 have a connections with the occurrence and development of Wilms' tumor.2,The expressions of SOCS-1 mRNA and SOCS-3mRNA in UH were significantly lower than those in FH which illustrate the lower expressions of SOCS-1 mRNA and SOCS-3 may have a connections with the prognosis of Wilms' tumor.Significance In recent years, the researches about the expression of SOCS in cancers have caused much concerns,and the expression descended in many cancers.In our country we firstly researched the expressions of mRNA and protein of the SOCS-1 and SOCS-3,and the relation between the expressions and the different pathological classifications.The results consisitents with findings of the international.At the same time,we provide the theoretical basises and a new pathway for the gene therapy of Wilms' tumor.
