Background:Ulcerative colitis(UC)is defined as a diffuse non-specific inflammatory bowel disease(IBD)of unknown cause that continuously affects the colonic mucosa proximal from the rectum and often forms erosions and/or ulcers with possible complication in intestine and other organs.The cause of UC has not yet to be ascertained,but it is considered to develop as the result of abnormal immune response to host intestinal mucosa and the gut microbiota changes caused by environmental factors such as diet changes and infection in genetically susceptible individuals.And the disorder of the balance of Thl/Th2 type cell is a potential mechanism to alter the disease course of patients with ulcerative colitis.Although there are many western therapeutic methods available for the treatment nowadays mainly by blocking inflammation and regulating immune function,even biologics and sometimes operation,it still can not meet clinical needs for the disease.Due to insufficient relief of symptoms and maintenance remission,the recurrence rate and frequent intolerable side effects,it is difficult to maintain long term treatment for the patients with UC.While the traditional Chinese medicine can alleviate the clinical symptoms effectively and improve quality of life of UC patients,with advantages of a diversity of multi-target and less adverse reaction,having future prospects development for patients with ulcerative colitis.OBJECTIVETo determine the efficiency of the herb compound Yiqifuyuan decoction for the treatment of 2,4,6-trinitrobenzene sulfonic acid(TNBS)/ethanol-induced ulcerative colitis rats,and to explore the potential influence on Th1/Th2 type cells balance.METHODSAll rats were randomly divided into six groups:normal control group;model group;sulfasalazine oral treatment group;and low,medium,and high-dose Yiqifuyuan decoction groups(10 rats each).Except normal control groups all rats with ulcerative colitis were induced with 2,4,6-trinitrobenzenesulfonicacid(TNBS).From the day 3 after establishing models,different drugs were given to the corresponding groups by means of intragastric,and normal saline were given to the rats of normal and model group once a day at the same time for 14 days.The general condition,disease activity index(DAI),Colon mucosal damage index(CMDI)and histological injury score(HIS)of rats were evaluated after 14 days,and the content of interleukin-4(IL-4),Interferon-gamma(INF-?)and Tumor necrosis factor a(TNF-?)in peripheral blood were measured by ELISA respectively,to compare the difference among groups with different intervention.RESULTSCompared with rats in the model group,the DAI,CMD,HIS and the content of TNF-? and INF-?,CD4+T lymphocyte cell and the CD4+/CD8+ ratio in ulcerative colitis rats treated by Yiqifuyuan decoction and sulfasalazine were remarkably reduced(P<0.05).Meanwhile the serum IL-4 content,CD8+T lymphocyte cell,CD4+CD25+T lymphocyte cell,and the CD4+/CD8+ ratio of the model group were significantly lower(P<0.05)than that in the normal group,while that of the Yiqifuyuan decoction groups were significantly improved(P<0.05).CONCLUSIONThe research suggests that the treatment with Yiqifuyuan decoction is effective and has significant effects on pro-inflammatory cytokines and inflammatory cytokines suppression in ulcerative colitis rats induced by TNBS by regulating the Th1/Th2 type cells balance.
