| Part Ⅰ The Clinical Efficacy Research of Compound Sophorae Decoction in Treating Ulcerative ColitisOBJECTIVE: To identify the composition of Compound Sophorae Decoction and to study the clinical therapeutic effect of Compound Sophorae Decoction.METHODS: The ingredients of Compound Sophorae Decoction were determined.Matrine,Oxymatrine,Notoginsenoside R1,Ginsenoside Rb1,Gallic acid,Indigo,Indirubin,Glycyrrhizic acid and Liquiritin in components of Compound Sophorae Decoction were identified by mass spectrometry.Bletilla striata(Thunb.)Reichb.f.was identified using thin layer chromatography(TLC)test.A retrospective study of the clinical efficacy of Compound Sophorae Decoction in the treatment of patients with ulcerative colitis was implemented.The symptoms of 184 cases of inpatients with UC who were admitted to the Department of Integrated Traditional Chinese and Western Medicine and Department of Gastroenterology of Union Hospital of Tongji Medical College of Huazhong University of Science and Technology were analyzed during January 2010 to December 2012,and the improvement of the two groups(western medicine combined with Compound Sophorae Decoction and western medicine)was compared after the comprehensive treatment.RESULTS: Compound sophorae decoction was composed of Sophora flavescens Ait.,Sanguisorba officinalis L.,Indigo Naturalis,Bletilla striata(Thunb.)Reichb.f.,Glycyrrhiza uralensis Fisch.and panax notoginseng(Burk.)F.H.Chen.Retrospective analysis showed that Western medicine combined with Compound Sophorae Decoction was better than Western medicine in the treatment of patients with ulcerative colitis.CONCLUSION: When combined with western medicine,Compound Sophorae Decoction plays a significant synergistic effect in the treatment of ulcerative colitis.Part II The Effect of Compound Sophorae Decoction on regulating Th17/Treg Cells Balance in DSS-induced Colitis in MiceOBJECTIVE: To study the mechanism of Compound Sophorae Decoction in the treatment of dextran sodium sulfate(DSS)-induced colitis in mouse.METHODS: A mouse model of ulcerative colitis was administrated with DSS.The colitis mice were treated with Compound Sophorae Decoction and mesalazine.The stool,activity,and body weight of the mice were observed during experiment to assess the disease activity of the mice.After the treatment of two drugs,HE sections of the mouse colon tissues were observed to evaluate the pathological damage of the colon of the mice;the concentration of IL-1β,TNF-α,IL-17 A,IL-10,IL-6,TGF-β of the colon tissues of the mice were detected by ELISA;WB and IHC were used to detect the expression of phosphorylated NF-κB p65,STAT3 and phosphorylated STAT3 in colon tissue of mice.The percentage of Th17 and Treg cells in spleen and mesenteric lymph nodes were detected by flow cytometry.The expression levels of ROR-γt and Foxp3 in colon tissues were detected by PCR.RESULTS: After administrated with DSS,the activity and body weight of mice were significantly decreased,and blood in stools were observed.The colon tissue of the mice had congestion and ulcers.Compared with the normal group,the levels of inflammatory factors IL-1β,TNF-α,phospho-NF-κB p65 in the model group were significantly increased;the proportions of Th17 and Treg cells were increased,and the expressions of IL-17 A,IL-10,ROR-γt and Foxp3 were all increased;the expression of STAT3 and IL-6 were increased in the model group,while the expression of TGF-β was decreased.The symptoms and pathological damage of mice treated with Compound Sophorae Decoction and mesalazine were improved compared with model group.The expression of inflammatory factors IL-1β,TNF-α,phosphorylated NF-κB p65 were decreased;the differentiation of Th17 cells were inhibited,but the differentiation of Treg cells were increased.CONCLUSION: Compound Sophorae Decoction can regulate the immune function of DSS-induced colitis in mice by regulating the balance of Th17/Treg cells,and improve the symptoms and the pathological damage of mice with colitis. |
