Research background and objectives:Cervical cancer is currently the only known cause of etiology.It can take effective measures to prevent malignancy from occurring.TCT and HPV tests are now mostly used for cervical cancer screening.HPV tests are divided into HPV-DNA and HPV types.These two detection methods can only detect whether the body is infected with HPV,high-risk HPV cervical infection and HPV oncogene E6/E7 integration into the host cell,and the expression of the oncoprotein E6/E7 protein is a necessary condition for the progression of cervical lesions.While the detection of HPV E6/E7 expression compared with the simple detection of DNA,this method can better reflect the HPV oncogene integration into the host cell to play a carcinogenic effect,suggesting that the virus activity and cancer risk,can effectively reduce the transient HPV Detection of infection.This article aims to study the different levels of cervical lesions in the HPV16/18 E6,HPV16 E7 protein positive expression,and then to explore the application of HPV16/18 E6,HPV16 E7 protein in cervical cancer screening.Research methods:Random number method has used to select 10 paraffin-embedded specimens from patients with chronic cervicitis who are hospitalized in First Affiliated Hospital of Jilin University from 2014 to 2017,20 specimens from with CINI grade patients,40 specimens from CINII-III patients,and 10 specimens from patients with cervical squamous cell carcinoma.Immunohistochemical SP method has used to detect the positive expression of HPV16/18 E6 and HPV16 E7 protein in different grades of cervical lesions.Chi-square analysis is used to compare the positive rate of each group,and the correlation of E6/E7 protein expression is calculated by Spearman rankcorrelation analysis.P < 0.05 is statistically significant for differential study.Research results:1.HPV16/18 E6 protein,HPV16 E7 protein is mainly expressed in the nucleus,and the cytoplasm is also partially expressed.2.(1)HPV16/18 E6 protein was found in different cervical pathological tissues,and the positive expression rate was 20.0%,40.0%,65.0% and 80.0%,respectively,and the difference was statistically significant(?2=11.370,P=0.001).The positive rate of HPV16/18E6 protein increased with the severity of cervical lesions.Through Spearman correlation analysis between the four groups,it was found that the positive correlation between HPV16/18E6 positive expression and cervical lesion was positively correlated(r=0.410,P=0.000),indicating that the higher the degree of cervical lesions,the stronger the expression of HPV16/18 E6 was.(2)HPV16 E7 protein was found to be 10.0%,40.0%,67.5% and 90.0%respectively in cervical pathological tissues at different levels of cervical pathology.The difference was statistically significant(?2=17.524,P=0.000).The positive rate of HPV16E7 protein increased with the severity of cervical lesions.Through Spearman correlation analysis between the four groups,it was found that the positive expression of HPV16E7 was positively correlated with the degree of cervical lesion(r=0.506,P=0.000),indicating that the higher the degree of cervical lesion,the stronger the expression of HPV16E7 was.3,(1)In groups of five different cytology cervical organization,group,in comparison with HPV16/18 E6 positive expression in positive expression rate was21.2%,40.0%,62.5%,69.0%,100%,the difference is statistically significant(?2=24.376,P = 0.001).The positive rate of HP16/18E6 protein increased with the increase of TCT level in the cervix.Through Spearman correlation analysis between the five groups,it was found that HPV16/18 E6 protein positive expression was positively correlated with cervical TCT grading(r=0.378,P=0.01),indicating that the higher the cervical TCT level,the stronger the expression of HPV16/18 E6 protein was.(2)In the five groups,the positive expression rate of HPV16 E7 was 7.1%,40.0%,62.5%,76.2% and 100%,respectively,and the difference was statistically significant.Meaning(?2= 25.565,P=0.000).The positive rate of HPV16E7 protein increased with the increase of TCT level in the cervix.Through Spearman correlation analysis between the five groups,it was found that HPV16 E7 protein positive expression was positively correlated with TCT grading(r=0.514,P=0.000),indicating that the higher the cervical TCT level,the stronger the expression of HPV16 E7 protein was.4,Comparison group HPV16 /18 E6 protein,HPV16 E7 protein expression,in45 cases of HPV16 E7 protein expression is also positive,44 cases of HPV16/18 E6 protein positive expression in cervical tissue,through Spearman analysis,according to the results of the two proteins expression in cervical tissues were positively correlated(r = 0.791,P = 0.000).Research conclusions:1.The positive expression rate of HPV16/18 E6 protein and HPV16 E7 protein increased with the severity of cervical lesions,and the positive expression rate and expression intensity were positively correlated with the level of cervical lesions.The positive rate and intensity of expression in cervical cancer were significantly higher than those in chronic cervicitis and CIN groups.2.HPV 16/18 E6 protein,HPV16 E7 protein in different TCT cytology results,with the increase in cervical TCT grades,the expressions rate increased.3.HPV16/18 E6 protein,HPV16 E7 protein assay,with TCT assay for screening of Cervical Diseases,helping to predict the degree of disease progression and postoperative follow-up.
