| BackgroudCervical carcinoma (CC) is one of the most common cancers and a leading cause of death among women worldwide. The incidence of developing countries is significantly higher than developed countries1.Infection with high-risk HPV is a necessary-but not sufficient-cause for developing cervical carcinoma. Studies have found that 60% of low-grade squamous intraepithelial lesions (LSIL) can be naturally dissipated,5-10% of LSIL can develop into high-grade squamous of intraepithelial lesions (HSIL), only less than 1% of LSIL will develop into cervical carcinoma2. In the course of development of cervical carcinoma, wnt/β-catenin signaling pathway as a "second hit" leads the normal cells with HPV infected develop into HPV immortalized keratinocytes. It will be advantageous of improve the therapeutic program to aim directly at the etiological factor of the cervical carcinoma, and provide the forceful theoretical support for the clinical therapy.ObjectiveTo investigate the expression of secreted frizzled related protein-1 (sFRP-1), Wnt-1 in patients with cervical intraepithelial neoplasia and cervical carcinoma.MethodsCervical intraepithelial neoplasia(CIN) and squamous cervical carcinoma tissue (SCC) were picked from 80 patients from January 2007 to May 2009 who underwent surgery in our department, and none of them received preoperative radiotherapy, chemotherapy and other treatments. We used 14 normal cervical tissue picked from patients who suffered from myoma and underwent hysterectomy. The cervical intraepithelial neoplasia include 20 LSIL and 20 HSIL. sFRP-1 and Wnt-1 were detected by immunohistochemistry (SABC) in normal cervical tissue(picked from myoma),LSIL(including CIN I),HSIL (including CINⅡ,CINⅢand cervical carcinoma in suit) and cervical carcinoma, at the same time reverse transcription polymerase chain reaction(RT-PCR) detects the expression of sFRP-1,Wnt-1 mRNA in normal cervical tissueand,CIN and SCC.Results1. The sFRP-1 protein located in the membrane and cytoplasm, which appears brownish yellow granules. All the normal cervical epithelial tissue were positive staining and the sFRP-1 protein strong expressed in the basal layer and the most areas of acantholysis. The positive rate of the sFRP-1 protein in the CIN tissues is 75%, and the sFRP-1 protein weakly expressed in CIN tissues. The positive rate of the sFRP-1 protein in the SCC tissues is 40%, and the sFRP-1 protein expressed in the SCC more weakly. The expression of sFRP-1 protein is gradually weakened (P<0.05).2. The Wnt-1 protein located in the cytoplasm and some expressed in the membrane, which appears brown-yellow granules. The positive rate of normal cervical epithelial tissues was 21.4%, at the same times, the Wnt-1 protein expressed in the basal layer and grass-roots weakly.The positive expression rate of the Wnt-1 protein in the CIN tissues was 80%. The positive rate of the Wnt-1 protein in the SCC tissue was 100%, and the Wnt-1 protein expressed in the SCC tissues strongly. The expression of Wnt-1 protein is gradually increased(P<0.05).3. The expression of sFRP-1 mRNA in the SCC tissue didn't express or weakly expressed,while expressed in the normal cervical tissue significantly(x2= 15.556, P <0.05).4. The expression of WNT-1 mRNA in the normal cervical tissue didn't express or weakly expressed,while expressed in the SCC tissue significantly(x2= 9.956, P <0.05).Conclusions1. The expression of sFRP-1 protein in cervical cancer is significantly lower than HSIL and LSIL, while LSIL lower than normal cervical tissue.2. The expression of Wnt-1 protein in cervical cancer is significantly higher than HSIL and LSIL,while HSIL higher than LSIL, LSIL higher than normal cervical tissue.3. The lower expression of sFRP-1, over expression of Wnt-1 play a important role in the development from cervical intraepithelial neoplasia to cervical carcinoma. |
