Association Analysis Between Polymorphisms Of FADS3 Gene And Coronary Heart Disease

Research purpose We investigated the association of single nucleotide polymorphisms(SNPs)of rs1000778 in the fatty acid desaturase-3(FADS3)gene with coronary artery diease(CAD)and severity of disease in a case-control study and evaluated the possible influence on the lipid concentration in the two groups respectively.Research methods A total of 397 patients in Department of Cardiology,Qingdao Municipal Hospital,were selected from March 2016 to November 2017,including 243 patients in coronary heart disease group and 136 patients in control group.All patients in coronary heart disease group were confirmed by coronary angiography.The patients in the control group were confirmed by coronary angiography to be nearly smooth or underwent coronary CT angiography without stenosis.Patients with CAD were subclassified into 3subgroups(single-,double-and triple-vessel disease)based on the number of stenotic coronary arteries.The two groups were recorded with age,sex,history of smoking,history of hypertension and diabates mellitus,blood lipids,and so on.The venous blood was collected 5 ml and the peripheral white blood cell DNA was extracted with the consent of the patients.At the same time,the allel gene and genotypes of rs1000778 polymorphic loci of FADS3 gene were detected by Agena Mass Array DNA mass spectrometry system.Thefrequenciesof allel gene and genotypes were compared between the two groups and analyse the clinical characteristics of different genotype.SPSS23.0 software was used for statistical analysis.The measurement data are expressed by means±standard deviation and compared by the Independent-Samples T Test,and the qualitative data is expressed as percetages.Qualitative data were analyzed with the chi-square test.logistic regression was used to calculate the odds ratio(OR)and 95% Confidence interval(CI).According to the results to evaluated the association of rs1000778 single nucleotide polymorphisms(SNPs)in the fatty acid desaturase3(FADS3)with coronary artery disease and severity of disease.P<0.05 was considered to be statistically significant.Results Compared with control groups,patients with CAD were older(66.57±11.17P=0.001)and had higher percentage of male(65%vs47%,P=0.01)and have highe r i-ncidence rate of diabates mellitus.However,there was no statistical difference on history of smoking and lipid concentration.Based on coronary angiography,68(28.0%)patient in the CAD Group had single-vessel disease,60(24.7%)had double vessel disease and 115(47.3%)had triple-vessel disease.The minor allele(A)fre quency of rs1000778 was 0.1886,and the the genotype distribution in our study s-ubjects showed no deviation from the Hardy–Weinberg equilibrium(P >0.05).T here was no significant difference in the distribution of the genotype(P=0.292)and alleles(P=0.892)in the case group and the control group,We also did not find that the AG genotype(AGvs AA:OR=2.425,95%CI:0.763-7.714 P=0.1333)and GG genotype(GGvs AA:OR=2.026,95%CI:0.661-6.211 P=0.217)were associ ated with the risk of coronary heart disease.No association was fou-nd between the genotype and the susceptibility to coronary heart disease under the dominant model(GGvs GA+AA:OR=1.088,95%CI:0.698-1.696 P=0.71)and the recessive model(AAvs GA+GG:OR=2.143,95%CI 0.705-6.512,P=0.179).We assumed that different genotypes were independent of the number of diseased coronary art eries,obtained P(0.625)> 0.05 by chi-square test.We did not rej-ect the origin al hypothesis,and then we use an ordered logical regression.The results showe d that the coefficient of genotypic variables was not statistically sig-nificant(P=0.140,P=0.287),indicating that there was no association between the rs1000778 single nucleotide polymorphism and the severity of coronary heart disease.And n o association were observed between rs1000778 genotype and total cholesterol,tri glyceride,low density lipoprotein cholesterol and high density lipop-rotein choleste rol levels.Conclusion Our results suggest that the rs1000778 polymorphism of FADS3 gene was n ot associated with coronary heart disease and severity of coronary artery disease in Qingdao Han population,and the rs1000778 genotypes was not associated wit h lipid concentration.
